    Title: 發展並應用高特異性alcohol dehydrogenase gene (type III) primer快速檢測醋酸菌

    Abstract: 本研究利用已發表之AAB (acetic acid bacteria) type III adh sequence 設計 primer ADH III L187 / ADH III R500與AAB之reference strains進行PCR反應,可產生 313 bp的PCR產物。其他如Bacillus cereus, Yersinia enterocolitica、Salmonella enterica subsp.、Lactobacillus amylovorus、Pseudomonas aeruginosa及E.coli等非醋酸菌之菌株,則無任何 PCR 產物生成。此外,本實驗共篩選出75 株疑似醋酸菌,經 ADH III L187 / ADH III R500 增幅後,可產生313 bp PCR產物有 41 株,進一步應用Trcek et al.發表之alcohol dehydrogenase gene ( type I ) primer檢測AAB。結果顯示所分析之AAB菌株皆為 Acetobacter spp.,由此顯示 ADH III L187 / ADH III R500 primer對於醋酸菌具有高特異性。
