    Abstract: 本研究的目的在解決家禽霍亂疫苗保護力不佳的問題,藉由研發一新型的重組次單位疫苗,改善家禽霍亂疫苗的效力。利用二維電泳分離抗原,配合以不同保護力疫苗所免疫的抗血清進行免疫染色,鑑定菌體中的重要蛋白質抗原,再進而利用這些免疫原建構重組蛋白,研發次單位疫苗。
    The aim of this study is to develop a novel recombinant subunit vaccine to improve the efficacy of vaccine against fowl cholera. The study is proceeded by identify high immunogenic proteinaceous fowl cholera-specific antigens on Pasteurella multocida by performing immunoblotting after two-dimensional polyacrylamide gel electrophoresis (2D PAGE). The important immunogenetic proteins are recognized by antibodies of mice immunized with high-protective vaccine but not by sera from mice immunized with low-efficacy vaccine. Those proteinaceous immunogen candidates are identified by subjected to trypsin digestion followed by peptide mass fingerprinting and GeneBank search. In the second year, these genes will then be cloned and transformed to Escherichia coli for protein expression. In the third year, vaccine are prepared with these recombinant proteins and their efficacy are examined by challenge experiments in mice.
