摘要: | 本研究分別探討兩種水果第一部分以 常見水果 DL為研究對象 分開收集新鮮殼 (DL fresh hull, DLH F)與籽 (DL fresh seed, DLS F)及烘焙過的殼 (DL dried hull, DLH D)與籽 (DL dried seed , DLS D)利用乙醇和水 萃取進行抗氧化、抗發炎、預防光老化 及 DNA修復等試驗效能探討。無論是DL之 乾品或鮮品皆 具有清除 DPPH·和 ABTS· +能力,且 DLH D水萃物 (DLH DW)可以抑制人類皮膚角質細胞 (human skin immortalized keratinocyte, HaCaT)內活性氧物質(reactive oxygen species, ROS)及超氧陰離子 (superoxide, O2· 並提高穀胱甘肽之含量,同時透過增 加 超氧化物歧化酶 (superoxide dismutase, SOD)、 穀胱甘肽過氧化物酶 (glutathione peroxidase, GPx)及過氧化氫酶 (catalase, CAT)而達到抗氧化之功效。另外, DLH DW可以透過抑制 環氧合酶 ccyclooxygenase 2, COX 2)與 一氧化氮合 成 酶 (nitric oxide synthases, iNOS)之基因表現進而降低 腫瘤壞死因子、介白素 (interleukin 6, IL 6)、介白素之分泌。而在老化試驗中, DLH DW促進 膠原蛋白 (collagen)生成及 抑制玻
尿酸酶分解 並藉由提升修復因子的表現而預防因紫外線引起 之光化產物 嘧
啶二聚體 (cyclobutane pyrimidine dimer, CPD)及二聚體 (6 4 pyrimidine pyrimidine photoproducts, 6 4PP)之生成 。由本研究試驗結果得知,DL萃取物以 DLH DW抗氧化效果最佳,且同時具有抗發炎及預防光老化等效能。 因此期許 DLH DW之抗皺與修復皮膚之作用 未來可以應用於 化妝品的活性成分。 The research was included two fruits. The first part was the common fruit- DL. The fresh hull (DLH-F), fresh seed (DLS-F), dried hull (DLH-D), and dried seed (DLS-D) were collected separately. The samples were extracted with ethanol and water and the effects of anti-oxidation, anti-inflammation, anti-photoaging and DNA repair were evaluated.
DL extracts showed anti-oxidation effects by DPPH· and ABTS·+. DLH-D water extract (DLH-DW) could inhibit reactive oxygen species (ROS) and superoxide (O2·-) and increase glutathione (GSH) content and antioxidant enzymes in HaCaT cells. DLH-DW also inhibited tumor necrosis factor (TNF-a), interleukin IL-6, interleukin IL-1b by decreasing the gene expression of cyclooxygenase-2 (COX-2) and nitric oxide synthases (iNOS). Besides, DLH-DW could promote collagen production, degrade hyaluronidase, reduce the photochemical products cyclobutane pyrimidine dimer (CPDs) and 6-4 pyrimidine photoproducts (6-4PP) induced by UVB irradiation through regulating DNA repair factors.
In this study, DLH-DW showed excellent anti-oxidation effect in DL extracts and it also had anti-inflammation and photoaging prevention effects. Therefore, DLH-DW may apply in cosmetic ingredient in the future. |
