摘要: | 丹參(Salvia miltiorrhiza),為唇形科(Lamiaceae)植物,其根部主要含有親酯性的丹參酮及親水性的丹參酚酸衍生物。其中,丹參酮IIA及丹參酚酸B各為抗癌、保護心血管、降低血糖及抗氧化的活性指標成分。本研究探討本土種植丹參,丹參酚酸 B 與丹參酮IIA含量以及抗淋巴腫瘤細胞功效。以高效能液相層析法分析,丹參乙醇及水萃取物,所含的丹參酮 IIA 及丹參酚酸 B 的含量;以 MTT 方法測試丹參酮 IIA 、丹參酚酸 B 以及丹參萃取物對淋巴腫瘤細胞存活率的影響。結果顯示,丹參乙醇萃取物與水萃取物中丹參酚酸 B含量均大於5%以上,以丹參70%乙醇萃取物 (SmE70)中丹參酚酸 B含量最高,約8.6%。而丹參乙醇萃取物中丹參酮 IIA含量大於0.2%以上,以丹參70%及95%乙醇萃取物 (SmE70及SmE95) 含量最高,約0.6%,丹參水萃取物(SmW)只有0.03%。在 MTT 試驗中,丹參酚酸B 對細胞沒有毒性,丹參酮 IIA 在濃度0.5 g / ml 以上對淋巴腫瘤細胞具有毒性;在萃取物中,SmE50 、SmE70及 SmE95 在 濃度50 g/ml時影響淋巴腫瘤細胞存活率,約79.3%、 55.6%及59.5%;而 SmW 對細胞不具有毒性作用。 Salvia miltiorrhiza is a plant belonging to the family Lamiaceae. Its roots mainly contain the lipophilic tanshinone and hydrophilic salvianolic acid derivatives. Among them, tanshinone IIA and salvianolic acid B are index compounds of anti-cancer, cardiovascular protection, lower blood glucose and antioxidant activity. This study was to explore the salvianolic acid B and tanshinone IIA levels and anti-tumor activity of the local planting, S. miltiorrhiza local planted. The contents of tanshinone IIA and salvianolic acid B in the ethanolic and aqueous extracts from S. miltiorrhiza were determined by high performance liquid chromatography analysis. Effects of tanshinone IIA, salvianolic acid B and the ethanolic and aqueous extracts from S. miltiorrhiza on cell viability were measured by MTT method. The results showed that the contents of salvianolic acid B in the ethanolic and aqueous extracts from S. miltiorrhiza were more than 5%, and the 70% ethanolic extract from S.miltiorrhiza (SmE70) contained the highest content of salvianolic acid B, about 8.6%. Furthermore, the contents of tanshinone IIA were more than 0.2% in the ethanolic extracts from S. miltiorrhiza. The contents of 70% and 95% ethanolic extracts from S. miltiorrhiza (SmE70 and SmE95) were the highest, about 0.6%, while the content of the aqueous extracts from S. miltiorrhiza (SmW) was only 0.03%. In the MTT assay, salvianolic acid B did not affect cells viability, and tanshinone IIA was toxic to lymphoma cells at a concentration of 0.5 g / ml. These extracts from S. miltiorrhiza, SmE50, SmE70 and SmE95 at 50 g / ml were toxic to lymphoma cells, the survival rates were about 79.3%, 55.6% and 59.5%, respectively; however, SmW had no toxic to the cells. |
