本研究目的在於探討膳食蛋白質缺乏對於大鼠鐵代謝的影響。採用六週大S.D. 雄鼠10 隻,分成兩組:正常控制組(N 組)與蛋白質缺乏組(0% protein,PF 組),每組各5 隻,飼養8 天後犧牲,取血液與肝臟分析鐵代謝相關指標。結果顯示,PF 組體重與血漿白蛋白顯著降低,血紅素與控制組相當,血球比容顯著高於C 組,肝鐵濃度、鐵蛋白(ferritin)量、粒線體aconitase (m-ACO)酵素量分別為C 組的1.8 倍、9 倍、1.7 倍,肝鐵顯著堆積,且鐵儲存指標(ferritin)與功能性指標(m-ACO)均顯著增加;肝臟鐵調控蛋白IRPs 之RNA 結合活性,IRP1 不受蛋白質缺乏的影響,IRP2 於PF 組顯著降低。結論:短期蛋白質缺乏,降低IRP2 活性,顯著增加肝鐵濃度與鐵蛋白量,導致肝鐵堆積。 The aim of this study was to investigate the effect of dietary protein depletion on the iron metabolism in rats. 10 male SD rats (6-wks-old) were assigned to 2 groups (n=5), normal control (N) and protein-free group (PF). After 8 d feeding, all animals were sacrificed by carbon dioxide asphyxiation. Blood and liver were collected for analysis of iron-associated parameters. Our results showed that PF group had significantly lower body weight and serum albumin compared with N group. Hb level had no difference between 2 groups, but Hct was higher in PF group. The hepatic iron, ferritin, and mACO in PF goup were 1.8, 9 and 1.7 fold, respectively. The IRP2 activity was significantly decreased in PF group. In summary, protein depletion showed a decrease in IRP2 activity and higher iron and ferritin contents in liver. There was significantly hepatic iron accumulation in
protein-free group.
