A Bacillus subtilis F29-3 mutant defective in fengycin biosynthesis was isolated by Tn917 mutagenesis. Southern hybridization revealed that Tn917 in this mutant is inserted at a location outside of the chromosomal region containing the fengycin synthetase operon. Sequencing analysis showed that the transposon is inserted in an unknown gene fenY in the
nasA operon. The protein encoded by this gene potentially contains three phosphorylation sites and may be involved in the regulation fengycin synthesis. 枯草桿菌F29-3突變株的篩選是以Tn917轉位子進行突變作用所得到的,將這些突變株以南方墨點法分析,發現FE8突變株中Tn917轉位子所突變的位子不在豐原素peptide synthetase操縱組上。進一步以Tn917轉位子所突變的附近染色體序列作為探針篩選枯草桿菌F29-3基因庫,再分析DNA序列發現FE8突變株中Tn917轉位子所突變的位子是在與氮代謝有關的nasA操縱組附近的基因(fenY),FenY蛋白質序列分析發現含有三個磷酸化的位子,所以推測FenY可能參與在豐原素合成的調控上。
