    Title: 異源表達果糖轉移基因研究

    Abstract: 本計畫的目的是建立起異源表達的系統，以果糖轉移酵素基因為目標。我們選擇了大腸菌及pET系統，先將果糖轉移酵素的基因利用聚合酶鏈反應放大，再用限制酵素切割，最後利用連接酶構築於pET載體上，經轉殖入大腸菌後，以限制酵素切割檢驗及選出正確長度及正確方向的成功載體，最後以IPTG引發基因表達。我們成功在大腸菌中表達出果糖轉移酵素基因。
