人血小板衍生生長因子(PDGF)具有癒傷的生理活性,能夠增加粒狀組織的形成。在美國已有使用含0.01%人類重組血小板衍生生長因子的羧甲基纖維素鈉膠(NaCMC)於遠端神經性糖尿病潰瘍的治療。國外已有添加PDGF的化妝品出現,以幫助面部潰瘍的修復。本研究將人類PDGF-B的基因選殖於大腸桿菌載體,以酵素結合免疫吸附分析法(ELISA)及西方墨點法証實已經成功的將合成於細胞內的重組PDGF-B產物送出於培養基中。以super broth培養基與大腸桿菌菌株HB101於三角瓶振盪培養,可獲得最高的PDGF-B產率。 PDGF is a non-glycosylated protein that belongs to the family of dimeric cystine knot growth factors. There is considerable interest in the therapeutic potential of PDGF antagonists and agonists. The production of large quantities of PDGF is therefore desirable, and the simplest way to achieve this is by recombinant DNA technology. In this study, the human PDGFB gene was subcloned into an E. coli expression vector, pEZZ18, and the gene product was successfully secreted into growth medium as expected. The growth medium and E. coli strain that most efficiently overexpressed recombinant PDGFB were super broth and HB101, respectivrly, in the shake-flask studies.
