    摘要: 茵陳蒿為著名的肝病用藥,有消炎、抗氧化與抑菌的作用。在台灣常見的為綿茵陳( Artemisia capillaries )及北茵陳(Origanum vulgare L.)二個品種。綿茵陳有6,7-dimethyl esculetin、chlorogenic acid、caffeic acid、eugenol、scopoletin、2-ethtl phenol與o-cresol等成份;北茵陳有rosmrinic acid、thymol、carvacrol、luteolin、apigenin、caffeic acid、protocatechuic acid等成份。利用HPLC分析,以磷酸、水與氰甲烷為沖堤系統,完成綿茵陳及北茵陳市售品的分析圖譜與成份的定量。綿茵陳指標成份含量約為0.054~0.399 mg/g;北茵陳指標成份含量約為0.106~0.371 mg/g。包括DPPH清除自由基能力、 Trolox 當量抗氧化能力(TEAC)、清除超氧陰離子能力、抑制liposome脂質過氧化能力及酪胺酸酶的體外抗氧化試驗,結果均顯示,北茵陳優於綿茵陳。區段方面,DPPH自由基清除效應中,綿茵陳區段11、12、15、16、18在濃度為80 ppm時,清除率達85 %;北茵陳區段11、12、13、15在濃度為20 ppm時,清除率達80 %。當量抗氧化能力試驗中,綿茵陳區段12、19、20與北茵陳區段11、13、14、16清除效率皆達90%之樣品濃度分別為60 ppm及20 ppm。清除超氧陰離子試驗中,樣品濃度5 ppm時,綿茵陳區段19清除效率達50 %;北茵陳區段12清除效率達60 %。抑制 liposome 脂質過氧化能力試驗中,綿茵陳區段19與北茵陳區段9~12、15抑制效率皆達70 %之樣品濃度分別為80 ppm及60 ppm。在抑制B16-F10細胞分泌黑色素試驗方面綿茵陳無抑制效果;而北茵陳在濃度50 ppm時抑制率為11 %。北茵陳區段14、15、16之抑制率分別為29.8 %、22.9 %、37.9 %。在細胞毒性試驗中顯示北茵陳及其區段無細胞毒性。
    Artemisia Capillaris is a herb of the composite family. It reduces inflammation, antioxidant and antimicrobial activity. In Taiwan, Artemisia Capillaris and Origanum vulgare are often used as cure for jaundice. The former contains 6,7-dimethyl esculetin, capillarisin, chlorogenic acid, caffeic acid, eugenol, scopoletin, 2-ethtl phenol and o-cresol; the latter contains rosmrinic acid, thymol, carvacrol, apigenin, caffeic acid and protocatechuic acid. A simple method for simultaneous determination of these marker substances by high-performance liquid chromatography was developed. It was carried out by using a Mightysil RP-18 column with a gradient solvent system consisted of phosphoric acid and acetonitrile at 280 nm. Results showed that marker components of Artemisia Capillaris were 0.065~0.399 mg/g and those of Origanum vulgare were 0.106~0.371 mg/g. In antioxidant test, 2,2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging activity, trolox equivalent antioxidant capacity (TEAC), SOD radical scavenging activity and liposome methods of Origanum vulgare were better than Artemisiae Capillaris . Fractions 11、12、15、16 and 18 of Artemisiae Capillaris showed more than 90% scavenging activity when the sample concentration was 80ppm and fractions 11、12、13、 15 of Origanum vulgare showed more than 80% scavenging activity when the sample concentration was 20ppm in DPPH assay. In TEAC assay, fractions 12、19、20 of Artemisiae Capillaris and fractions 11、13、14、16 of Origanum vulgare present 90% scavenging activity when the sample concentration were 60ppm and 20ppm. In SOD-like assay, when the sample concentration was 5ppm, fraction 19 of Artemisiae Capillaris scavenging activity were 50%;fraction 19 of Origanum vulgare scavenging activity were 60%. In liposome method assay, Artemisiae Capillaris fraction 19 and Origanum vulgare fraction 9~12 and 15 inhibition activity more then 70% when the sample concentration were 80ppm and 60ppm. In cell test, we found that Artemisia capillaries could not inhibit the B16F10 proliferation melanoma. On the other hand, the inhibition activity was 11% when Origanum vulgare concentration was 50 ppm. Besides, the inhibition activity of Origanum vulgare fraction 14、15 and 16 were 29.8 %、22.9 % and 37.9 %. MTT assay demonstrated that the Origanum vulgare L. and its fractions had not toxicity.
