    摘要: 碳水化合物在釵h自然界物質中佔有重要的地位,在一些中草藥中具有有效成份的弁遄A所以要選擇一種簡單、快速、可靠及具有再現性的方法,以毛細管電泳電化學偵測法(CE-ECD)為基礎,針對傳統中草藥中碳水化合物的偵測。冬蟲夏草中甘露醇具有一定的藥效成份,對冬蟲夏草有很大的影響,所以本實驗利用毛細管電泳電化學偵測法,針對工作電極電位、電解液的濃度、電解液的組成和分離電壓及注射時間的影響進行研究。在這個方法,不需要進行衍生,所以不易受到其他物質的干擾。分離和偵測需要使用鹼性電解液環境,而且本實驗所使用毛細管較一般的長度短(20cm),內徑20μm,大大縮短分離時間及成本。利用電泳電解液的組成,在最佳分離情況下,很成奶擢鱁輕鴠_冬蟲夏草中甘露醇的成份,另一方面也可以來確認冬蟲夏草的品質的穩定。
    In this study, we developed a capillary electrophoresis(CE) separation combined with electrochemical detection(ECD) method for the determination of mannitols in Cordyceps militaris. A simple, fast, reliable and reproducible method based on capillary electrophoresis with electrochemical detection(CE-ECD), for the determination of carbohydrates in Chinese traditional drugs was described in this work. A copper microelectrode was used as working electrode. The effects of the potential of the working electrode, and concentration of running buffer, electrolytes composition of running buffer, separation voltage, and injection time on CE-ECD were investigated. In this method, both the separation and the detection required the use of alkaline electrolyte with 20cm long, 20μm i.d. fused silica capillary. With the optimized condition of separation and detection, the migration time of mannitol is 315(±7)s determination with dynamic range of 10.00mM to 0.10mM and of 0.05mM(LOD)limit of detection.
