    Title: 中草藥之生物活性探討及在化粧品之應用
    Studies on bioactivities and cosmetics application of the Chinese herbs extracts

    Abstract: 在中國,使用中草藥植物處理人類的疾病已有很長的歷史。然而,目前仍缺乏科學性的探討,輔以驗證其生物活性之有效性。
    本研究著重在中草藥植物萃取液之生物活性之探討與應用在化妝品配方之評估。由五十四種中草藥植物之粗萃液,透過「圓盤擴散」(Disc Diffusion)進行分析篩選,結果發現其中三種中草藥植物具有明顯的皮屑芽胞菌抑制作用。
    在生物活性之抑菌作用方面,結果顯示(1)PME、GCE及CFE針對皮屑芽胞菌之最小抑制濃度分別為3.125、6.250、6.250 mg/ml;(2)針對大腸桿菌之最小抑制濃度分別為0.781、0.390、0.195 mg/ml;(3)針對金黃色葡萄球菌之最小抑制濃度則分別為0.195、0.781、6.250 mg/ml。
    在清除自由基作用方面,運用DPPH、TEAC、Griess與Liposome等方法進行實驗。結果發現(1)PME、GCE及CFE在DPPH方面之SC50分別為1.09、1.20、1.06 mg/ml;(2)在TEAC方面之SC50 則分別為1.07、1.84、1.50 mg/ml;(3)在Griess方法分析方面SC50 分別為0.60、1.65、1.61 mg/ml;(4)在Liposome方法方面SC50則分別為0.69、0.64、0.57 mg/ml。(5)在清除超氧陰離子方面(SOD-like activity)GCE萃取液之活性為15885.42 U/mg。
    最後,將實驗結果實際運用在化妝品配方中,評估體外試驗結果與產品實際運用效果之差異性,結果顯示可提供商業上化妝品的良好應用。
    In China, it has been for a long time to make use of herbs for medical treatments of human diseases. However, effects on biological activity were not clearly proven for lack of scientic study.
    In this thesis, it emphasizes that how to study biological activity of herbs extracts and evaluate applications to cosmetics formula. Crude extracts of fifty-four species of herbs were screened by means of Disc Diffusion assay. It shows that three of them possess obvious inhibition effects upon Pityrosporum ovale.
    As for inhibition of biological activity, it shows that the minimum inhibition concentrations by PME, GCE, and CFE are (1) 3.125, 6.250, 6.250 mg/ml respectively for Pityrosporum ovale, (2) 0.781、0.390、0.195 mg/ml respectively for Escherichia coli, (3) 0.195, 0.781, 6.250 mg/ml respectively for Staphylococcus aureus.
    As for radical scavenging, DPPH, TEAC, Griess, and Liposome methods were conducted. It shows that the minimum inhibition concentrations by PME, GCE, and CFE are (1) 1.09, 1.20, 1.06 mg/ml respectively for DPPH SC50, (2) 1.07, 1.84, 1.50 mg/ml respectively for TEAC SC50, (3) 0.60, 1.65, 1.61 mg/ml respectively for Griess SC50, (4) 0.69, 0.64, 0.57 mg/ml respectively for Liposome SC50. Furthermore, the activity of GCE extracts is 15885.42 U/mg for superoxide radical scavenging activity.
    These experimental results were applied to cosmetics formula in order to compare results of in vitro tests with effects of practical cosmetics applications. Furthermore, it has been proven that the study is suitable for the development of commercial cosmetic products.
