    標題: 微乳液包覆制首烏萃取物之探討
    The Study on the Extract of Polygonum Multiflorum Thunberg Encapsulated in Microemulsion
    本研究以不同濃度乙醇溶液萃取制何首烏,並比較不同濃度乙醇萃取之制何首烏萃取物中指標成分的差異。本研究發現,以90%乙醇溶液萃取出的制首烏其指標成分二苯乙烯?,明顯高於50%乙醇浸提液萃取一倍。萃取之後以微乳液包覆制何首烏萃取物,比較Decyl Glucoside和Tween 80兩種界面活性劑製備含制首烏萃取物的微乳液差異,所備製的微乳液微包粒徑分別為14.9nm和94.0nm。分散係數分別為0.150與0.871。pH值分別為9.63與4.69。30天的安定性評估結果顯示,在25℃與45℃的保存下,粒徑大小較為穩定。但在4℃的保存條件下,粒徑大小明顯變化大。而在兩者酸鹼值上,Decyl Glucoside較偏鹼性約7~9之間,Tween 80的配方則多維持在4。在總量分析上Decyl Glucoside對指標成分有明顯的降解作用。而Tween 80則在第三十天時,不同保存溫度之微乳液指標成分總量仍維持在90%左右。以Tween微乳液進行皮膚吸收試驗。結果顯示,三重複分析之結果顯示指標成分的皮膚滯留量,分別各佔總量的16.25%、13.03%與9.64%。
    In this study, First-Polygonum multiflorum was extracted with different ethanol concentration. It was found that Stilbene glycosides, the index compomnent of Polygonum multiflorum, extracted with 90% ethanol solution was twice higher than the extract by 50%. After extraction, Polygonum multiflorum extract coated with microemulsion. Decyl Glucoside and Tween 80 were used as different surfactants to comparision. The result showed-the particle sizes were 14.9nm and 94.0nm, respectively. The dispersion coefficients are 0.150 and 0.871 . The pH values are 9.63 and 4.69. The 30-day stability evaluation results show that the particle size is relatively stable when stored at 25°C and 45°C. But under the storage conditions of 4℃, the particle size were getting larger under 4°C. The pH value of Decyl Glucoside microemulsion is around 7-9, and the formula of Tween 80 is mostly maintained at 4, the index compound in Decyl Glucoside formulation is a significant degraded . On the other hand, the index compound in Tween 80 formulation remained at about 90% after 30 days. The Tween 80 formulation was used for skin absorption experiment in triplicate, the results showed the index compound in skin were 16.25%, 13.03% and 9.64% of total.
