    Title: Diclofenac 的光解反應探討

    Abstract: 本實驗使用雙氯芬酸(Diclofenac)作為反應物,以低壓汞燈結合高效能液相層析儀(High Performance Liquid Chromatography, HPLC)測定光解前、後反應物及產物的濃度,計算藥物光解之動力學參數;再以中壓汞燈長時間照光並結合 HPLC 分析出光解後產物測定;最後以氣相層析質譜儀(Gas Chromatography-Mass Spectrophotometer, GC-MS)鑑定可能的光解產物結構,推論出 Diclofenac 可能的光解反應機構。本實驗中,使用 C-18 為HPLC分離管柱,最佳化分離條件以甲醇:水=75:25為移動相,流速 1.0 mL/min,在紫外光 280 nm 偵測。GC-MS之最佳化條件注射口溫度280℃,升溫程式從 100℃ 上升到 230℃,載流氣體(氦氣)的流速為 1.0 mL/min,電子撞擊游離電壓為 70 eV,四極柱溫度為 150℃。經由此次實驗推論出 Diclofenac 光解反應趨近於 0.4 級反應,在 25℃ 下,速率常數 k 為 1.34×10-6±8×10-8 ; 40℃ 下,k 為 1.56×10-6±3×10-8,光解活化能(Ea) = 7.654kJ/mol。以 GC/MS 鑑定光解的可能產物為 2-(Phenylamino)benzeneacetic acid 、Diclofenacmethyl ester 、4-Amino-9-fluorenone 、2-[(2,6-Dichlorophenyl)amino]benzeneethanol 、1-(2,6-Dichlorophenyl)indolin-2-one。
