    Title: 市售保健食品中甲硒胺酸和硒甲基硒半胱胺酸之測定

    Abstract: 本實驗製配 5 種硫醇(thiol)化合物薄膜 (2, 4-thiazolidinedione,2-mercapto-5-thiazoline, 2-mercapto-5-thiazoline , L-cysteine, thioglycolic acid),修飾金電極當工作電極。並裝入流動式電解槽 (flow cell)中,製造檢測器並連接高效率液相層析儀,測定甲硒胺酸 (L-selenomethionine) 和甲基硒代半胱胺酸 (Se-methylseleno-L-cysteine)之含量,探討出5 種硫醇(thiol)修飾金電極測定之靈敏度皆比未修飾金電極為高。5 種硫醇(thiol)修飾金電極靈敏度排序為 thioglycolic- acid> L-cysteine> 2,4-thiazolidinedione> 2-mercapto-5-thiazoline>2-mercapto-5-thiazoline。工作電極為鍍 0.06 mM 硫代羥乙酸(thioglycolic acid)修飾金電極感度最佳。層析 條件為移動相 甲醇 : 磷酸二氫鉀 比例為 10:90(V ∕ V),緩衝溶液(pH 3.86)為最適合條件,再進一步利用高效液相層析儀串連電化學偵測器串聯紫外可見光光譜儀(HighPerformance Liquid Chromatography – Electrochemical- Ultraviolet,HPLC-ECD-UV )找出甲硒胺酸滯留時間為3.597 分鐘、硒甲基硒半胱胺酸滯留時間為2.763 分鐘,結果找出TGA/Au 當工作電極的電化學偵測器感度是紫外可見光光譜的約1000 倍,並應用於 3種含 L-selenomethionine 及2 種含 Se-methylseleno-L-cysteine 的市售樣品。
