    Abstract: 本發明係有關於一種製備鄰位-羥基異黃酮的方法,其包含下列步驟:製備一含有融合基因之環狀重組質體,其中融合基因係具有CYP57B3基因之跨膜區?(transmembrane?domain,TMD)、CYP105D7基因與細胞色素還原?sCPR (cytochrome P450 reductase)基因;以及將環狀重組質體置於一適合的微生物表達系統中,並利用含有異黃酮之培養基培養微生物表達系統一作用時間;藉此,可提高鄰位-羥基異黃酮 (ortho-hydroxyisoflavones)的產量。 The invention relates to a method for manufacturing an ortho-hydroxyisoflavone. It comprises the steps of preparing a cyclic recombinant plasmid including a fusion gene, wherein the fusion gene comprises a transmembrane?domain (TMD) of CYP57B3 gene, CYP105D7 gene, and sCPR (cytochrome P450 reductase) gene; transforming the cyclic recombinant plasmid into a suitable microbial expression system; and incubating the microbial expression system in a medium containing isoflavone therein for a period of time. Accordingly, the yield of the ortho-hydroxyisoflavone can be improved.
