    Title: 紫色狼尾草熱水萃取物對於Streptozotocin誘導糖尿病小鼠血糖調控之影響
    Effect of Hot Water Extracts of Purple Napiergrass (Pennisetum purpureum Schumach) on Blood Sugar Regulation in Streptozotocin-Induced Diabetic Mice.

The hot air dried powder was extracted with 30 volumes of hot water for 10 minutes, and then concentrated under reduced pressure to prepare Pennisetum purpureum Schumach. extracts (PPE). In vitro test, determination of antioxidant components (total phenolic compound content and total flavonoid compound content), antioxidant test (seavenging DPPH radicals ability, trolox equivalent antioxidant capacity (TEAC), ferrous reducing antioxidant capacity (FRAC) and ferrous ion chelating ability), antiglycation test (inhibition of α-amylase, α-glucosidase and AGEs activity assay). In vivo test, 8-week-old male ICR mice were intraperitoneally injected with Streptozotocin (STZ) (i.p. 75 mg/kg BW) 3 weeks to induce diabetic mice. Divided into five groups : normal control group (Normal), diabetic control group (DM), low dose group (L), middle dose group (M) and high dose group (H) , The experimental group was tube feeding extracts 100, 250 and 500 mg/kg BW/day for 4 weeks, and free diet water during feeding. The results showed the extracts had antioxidant and antiglycation ability in vitro. In vivo, there was no difference in the antioxidant enzymes (catalase, SOD and GPX) in the liver. L group can down- regulate ox-LDL and increase HDL-C;M group can down-regulate ox-LDL;and H group has improved hyperglycemia, triglyceride and down-regulated ox-LDL caused by diabetes. Therefore, this study suggests that the diabetic mice tube feeding PPE had antioxidation and antiglycation ability in the H group, and the L and M group had antioxidant ability.

    Abstract: 本研究目的主要探討紫色狼尾草熱水萃取物是否對血糖具有調控之功效。紫色狼尾草熱風乾燥粉末以30倍體積的熱水萃取10分鐘後,再以減壓濃縮製備成紫色狼尾草萃取物。體外試驗檢,測抗氧化成分分析(總酚類物質及類黃酮物質含量)、抗氧化試驗(清除DPPH能力測定、總抗氧化能力(TEAC)測定、鐵還原力測定及螯合亞鐵離子能力測定)、抗糖化試驗(抑制α-amylase、glucosidase及AGEs活性測定)。體內試驗部分,以8週齡雄性ICR小鼠,腹腔注射Streptozotocin (STZ) (i.p. 75 mg/kg BW) 三週,誘發成糖尿病小鼠。再分成:正常對照組 (Normal)、糖尿病對照組 (DM)、低劑量組 (L)、中劑量組 (M) 及高劑量組 (H) 共五組。試驗組每天分別以口餵管餵食紫色狼尾草萃取物 100、250及500 mg/kg BW,試驗共進行4週,飼養期間採自由飲食飲水。結果顯示,體外試驗具有抗氧化及抗糖化的能力;體內試驗,各組在肝臟抗氧化酵素 (catalase、SOD及GPX) ,均無差異。L 組可以下調氧化型低密度脂蛋白及增加高密度脂蛋白膽固醇;M 組則可以下調氧化型低密度脂蛋白;H 組具改善糖尿病所造成的高血糖、三酸甘油酯及下調氧化型低密度脂蛋白。本實驗結果支持,糖尿病小鼠餵予紫色狼尾草熱水萃取物在高劑量組具有抗氧化及抗糖化之能力,則低、中劑量組具有抗氧化能力。
