    標題: 利用農業廢棄物萃取物進行抗氧化、抗發炎、抑制黑色素生成及DNA修復能力之效能評估

    摘要: 本研究以CFKH水萃及乙醇萃取物進行試驗,證實CFKHE之清除DPPH∙及ABTS∙+自由基能力、一氧化氮自由基、還原力、螯合鐵離子、總酚、總黃酮、總多醣含量,均優於CFKHW,再以細胞試驗進行毒性測試,針對人類皮膚角質株化細胞(HaCaT)及老鼠黑色素瘤細胞(B16F10),結果顯示兩者均無明顯的細胞毒性。因此本研究使用CFKHE進一步探討抗氧化、抗發炎、抑制黑素色生成及細胞修復等機制作用。在抗氧化試驗中,CFKHE能保護細胞免於受到H2O2誘發所造成的細胞內氧化壓力,同時提高Glutathione (GSH)含量及抗氧化酵素Superoxide dismutase (SOD)、Catalase和Glutathione peroxidase (GPx)的表現。在抑制黑色素生合成試驗中,以alpha-melanocyte stimulating hormone (α-MSH)誘導B16F10細胞的黑色素生成,發現CFKHE能抑制酪胺酸酶活性及降低黑色素的生成,並以免疫螢光試驗及西方墨點法驗證黑色素生合成相關因子。在HaCaT細胞的抗發炎試驗中,CFKHE能抑制lipopolysaccharide (LPS)所誘導之發炎因子Tumor necrosis factor alpha (TNF-α)、Interleukin 1 beta (IL-1β)、Interleukin 6 (IL-6)的生成。在DNA修復及保護能力試驗中,結果顯示HaCaT細胞經UVB照射後,CFKHE能促進傷口癒合(Wound healing)及降低免疫螢光UV光產物Cyclobutane pyrimidine dimer (CPD)含量。結果證實CFKHE具有抗氧化、抗發炎、抑制黑色素生合成及皮膚表皮細胞之DNA保護、修復能力,因此CFKHE具發展成為保養品添加劑的潛力。
    This study investigated the antioxidant DPPH free radical scavenging, ABTS free radical scavenging, reducing power, metal chelating activity, NO free radical scavenging, total phenol content, total flavonoids content, total Polysaccharides content, from CFKHW and CFKHE extraction, by cytotoxicity measurement by keratinocytes (HaCaT cells) and melanoma (B16F10 cells) and further study investigated anti-oxidant, anti-melanogenesis, anti-inflammation and cell repair properties.CFKHE also decreased cellular oxidative stress, increased intracellular GSH content and enhanced the antioxidant enzyme activities, such as superoxide dismutase (SOD), catalaseand glutathione peroxidase (GPx). CFKHE also decreased celluar tyrosinase and melanogenesis the immune fluorescent, western blot in melanocortin 1 receptor (MC1R), microphthalmia-associated transcription factor (MITF), tyrosinase, tyrosinase-related protein 2 (TRP-2) and tyrosinase-related protein 1 (TRP-1).In the anti-inflammation assay in CFKHE also decreased cellular inflammatory factors and UV induced cellular damage.This study demonstrated that CFKHE had antioxidant, anti- inflammation, protecting and repairing of DNA as well as decreasing inflammatory factors production. CFKHE had a potential to become novel health care products or food or cosmetic additives on whitening, antioxidant and anti-inflammatory.
