    Title: 虎杖對EB病毒溶裂循環的抑制效應
    Inhibitory effects of Epstein-Barr virus lytic cycle by Polygonum cuspidatum

    Abstract: 背景與目的:虎杖(Polygonum cuspidatum Sieb. Et Zucc),常見於民間使用之中草藥,它具有降低膽固醇、抗病毒、抗菌、抗腫瘤的活性。我們的研究目的主要是探討虎杖乙醇萃取物及其所含有的生物活性化合物,大黃素(emodin)與白黎蘆醇(resveratrol),對於抑制EB病毒溶裂循環的效應)。方法:以trypan blue分析細胞存活率,以西方墨點法,免疫螢光法及流式細胞儀分析EB病毒溶裂循環蛋白質的表現,以Real-time PCR分析病毒顆粒,以螢光素?分析病毒溶裂循環基因轉錄。結果:結果顯示虎杖乙醇萃取物對於P3HR1細胞的CC50為67.47 g/ml。虎杖乙醇萃取物、大黃素與白黎蘆醇分別在25 g/ml、9.37M及55M時具有抑制EB病毒顆粒量。虎杖乙醇萃取物濃度25g/ml 對病毒溶裂循環早期基因BRLF1及BZLF1轉錄抑制能力分別為77.29%及70.53%,大黃素在濃度9.37M的抑制能力分別為16.93%及31.17%;而白黎蘆醇在濃度6.25M抑制BRLF1及BZLF1轉錄能力分別為88.11%及92.04%。結論:本研究發現虎杖乙醇萃取物與其主要成份大黃素及白黎純具抑制EB病毒早期基因的表現,並且影響病毒裂蛋白質的表現,進一步抑制病毒的複製而達到抗病毒的效果。
