申請人於以往研究結果發現—日本沼蝦(Macrobrachium nipponense)應該可以作為監測淡水溪流中類雌性素(xeno-estrogen)的模式生物。E-screen assay 為採取水樣測定水中雌性素當量(estradiol equivalent concentration, EEQ)的常用方式。E-screen assay 主要有成本昂貴、水樣無法代表長時間的水體變化等問題。申請人為了延續以往的研究結果,擬以日本沼蝦為模式生物建立一套快速、便利、便宜且具生態意義的雌性素當量的測定方式。為建立測定方式,本計劃擬應用一年的時間進行兩部分的試驗,第一部分為建立雄性日本沼蝦與不同濃度的雌性素(17β-estradiol) 、 不同時間下卵黃前質蛋白(vitellogenon)誘發曲線—p-EEQ (estradiol equivalent concentration form pawn, p-EEQ);第二部分為確認p-EEQ 曲線之可行性,該部份將進行
臺灣五條溪流、中及下游,水樣與日本沼蝦的採樣。採回水樣經萃取後,以E-screen assay 方式來瞭解水樣類雌性素類環境荷爾蒙的當量濃度(EEQ), 同時並測定該水樣採集地點雄性日本沼蝦體內卵黃前質蛋白的濃度後計算該水體中之p-EEQ。比較五條溪流上、中及下游水體中類雌性素類(EEQ)的濃度與該處雄性日本沼蝦體內卵黃前質蛋白的濃度所得的p-EEQ 來判斷,雄性日本沼蝦體內卵黃前質蛋白變化是否可作為監測淡水溪流中類雌性素的生物指標。 A great deal of effect has been made on xeno-estrogen biomarker used in oriental river prawn (Macrobrachium nipponense). What seems to be lack, however, is field experiment. This paper is intended as an investigation of relationships between vitellogenin induction, xeno-estrogen biomarker in animal, and estradiol equivalency (EEQ) in the Taiwan River. In order to deep going research this subject we catch oriental river prawn from five rivers and measure concentration of vitellogenin in male prawn hepatopancreas, as well as use E-Screen assay measure EEQ in nature water. Hopefully, the results of this series research work will be useful as to monitor xeno-estrogen in freshwater in the future.
