    摘要: 背景和目的:順鉑(cisplatin,CP)是極為廣泛的癌症化學治療用藥,主要應用於治療頭頸部癌症、 食道癌、生殖器癌症、 子宮頸癌等。但也具有一定毒性,如:神經毒性、耳毒性、腎毒性 以及噁心嘔吐等,尤其以腎毒性為臨床使用上的主要限制。本實驗以不同條件進行白鳳菜(Gynura formosana,G) 水萃測試,最後採用熱水煮三小時做為動物實驗預防藥物,使用CP單一注射引發急性腎衰竭(Acute renal failure,ARF)評估在此模式中是否能對腎臟具有保護作用。方法:實驗使用9週齡C57BL/6品系雄性小鼠,適應一週後依樣品不同分成:空白組(施打saline 10mL/kg BW,Control組);誘導組(腹腔注射CP 20mg/kg BW,CP組);樣品組有白鳳菜高劑量組(GH,1 g/kg BW,以及白鳳菜低劑量組(GL,0.5 g/kg BW),實驗樣品連續管餵三週後,每組除了Control施打 Saline其餘皆施打CP進行誘導。於施打後72小時以CO2窒息犧牲,採集血清及組織。結果:白鳳菜萃出物以煮三小時總類黃酮含量最高,經由HPLC分析白鳳菜萃出物發現含有caffeic acid 與 Rutin,但是未見有明顯的Gallic acid 與 Quecertin成分;於心臟、睪丸、副睪脂、腹膜脂相對組織重量各組間並無顯著差異。但脾臟則C組顯著高於其他組別(P<0.05)。肝臟CP、GL組顯著低於C組 (P<0.05)。血清BUN、LDH中CP組顯著高於C組 (P<0.05)。GH組GPx活性顯著高於CP組(P<0.05)。高劑量白鳳菜水萃物對cisplatin 引起的急性腎臟GPX活性下降情形具有回復之作用。
    Background and purpose: Cisplatin (CP) is a very wide range of chemotherapeutic drugs, mainly used in the treatment of head and neck cancer, esophageal cancer, genital cancer, cervical cancer and other cancers. But also cause many side effects, such as: neurotoxicity, ototoxicity, renal toxicity, and nausea and vomiting, especially as the main limiting renal toxicity in clinical use. In this study, a single injection of CP cause acute renal failure (ARF), to Gynura formosana (G) immortality as a preventive medication, to assess whether the kidneys have a protective effect in this mode. Methods: Nine-week-old C57BL/6 strain male mice were divided into 4 groups according to different samples after 1 week of acclimation. Control group (C, administered playing saline 10mL/kg); treated group (CP, intraperitoneal injection of CP 20 mg/kg); sample groups were administered with aqueous extract 1 g/kg (GH) or 0.5 g/kg (GL) body weight. The mice were tube fed with aqueous extract or saline daily once for three weeks, then a single CP injection was given except C group. After 72 hours of CP injection, mice were sacrificed. Results: Gynura formosana extract for three hours has the highest levels of total flavonoids. Caffeic acid and Rutin was found by HPLC separation, howerver, no significant peak of Gallic acid and Quecertin was observed in the extract. Relative tissue weights showed no significant differences between the groups in heart, testes, EP, RE. Levels of serum BUN and LDH in CP group were significantly higher than that in group C (P <0.05). GPX activity of kidney in GH group was significantly higher than that in CP group (P <0.05). The acute decline of kidney GPX activity induced by CP injection could be restored with high dose of Gynura formosana extract.
