    Title: Imidazolium 側鏈對抗瘧疾藥物之微透析回收率的影響
    Effects of the Imidazolium Side Chains on the Microdialysis Recovery of Antimalarial Drugs

    Abstract: 微透析取樣技術,具有取樣量少、不影響生物之生理現象,因此可應用於活體動物之取樣,兼具簡單的樣本純化功能,現今已被廣泛地應用於各種生物樣本取樣分析中。然而許多具有藥理活性之物質,其高蛋白質結合率或親脂性高,以致回收率低,造成分析之困擾而不易對其分析物定量。 離子液體是在攝氏一百度下即可以呈現液態的有機鹽類,其中離子液體可做為溶劑,對許多種物質都有相當好的萃取表現。本實驗利用適當條件的咪唑類類離子液體,於體外微透析當中來有探討其側鏈對分析物的回收率影響。 本研究使用之抗瘧疾病藥物,其在治療效果與血中濃度關係具有監測之價值,故研究中利用適當條件之分析方法偵測微透析樣品,單一步驟同時定量五個典型抗瘧疾病藥物之微透析樣品9-Aminoacridine、Primaquine、Chloroquine、Quinacrine 與 Amodiaquine。高效液相層析-紫外光偵測器於 C-18 (25cm×4.6mm i.d.,5μm) 管柱下控制溫度 40℃,使用移動相acetonitrile:methanol:phosphate buffer (pH 6.6)=25:40:35(v/v%),流速 0.9mL/min,可於 35 分鐘內分離五種藥品,並使用 UV 波長 250nm 偵測定量。 微透析灌流液中添加 Imidazolium 類離子液體,控制微透析變因:不同側鍊離子液體、離子液體濃度、灌流液 pH 值、灌流速率等參數。結果指出,離子液體之碳鏈長度會影響微透析回收率,不同抗瘧藥物有明顯之回收率增幅,以[BMIm][Br]2.2mM 有最佳化結果。結果顯示導入離子液體於微透析技術中,能顯著地提升分析物之回收率,以達降低偵測極限濃度等更有利的分析條件。
    Microdialysis is a minimally-invasive sampling technique that is used for continuous measurement of free, unbound analyte concentrations in the extracellular fluid of virtually any tissue. Ionic liquid (IL) is a class of organic salts that can remain liquid state below 100 degree, and have recently been synthesized and applied in various fields.IL appears to be an ideal solvent in many extraction experiments, and could be immiscibility with water or organic solvents.Owing to these advantages, this study demonstrates the IL using on microdialysis for enhancing recovery of analytes. In our study.These drugs are also valuable for monitoring blood concentration. Therefore, it's advantageous to use suitable condition to determinate multiple antimalarial drug including 9-aminoacridine, primaquine, chloroquine, quinacrine and amodiaquine from microdialysis sample at once.The samples were separated on HPLC-UV by using Thermo Syncronis C-18 (25cm×4.6mm i.d., 5μm) column and mobile phase of acetonitrile methanol:phosphate buffer (pH 6.6) (25:40:35 (v/v)) in 0.9mL/min flow rate. All drugs were monitored and quantified by UV 250nm. Results of this study show different side chains of imidazolium ILs recovery increasing.Results of this experiment indicate that IL could significantly increase recovery of antimalarial drugs on microdialysis and accomplish the lower limitation of detection.
