    Title: 恆春半島特有植物萃取物之生物活性探討
    Evaluation of bioactivities of aqueous extracts from endemic plants in Hengchun Peninsula

    Abstract: 本研究主要探討台灣三種生長於恆春半島之熱帶海岸林植物: 棋盤腳(Barringtonia asiatica(L.)Kurz) (花、花柄)、白水木(Tournefortia argentea L. f.) (果實、葉、花)、石栗 (Aleurites moluccana ( L. ) Willd.) (葉、花) 的水萃物生物活性探討。 藉由測定清除DPPH 自由基、螯合亞鐵離子及TEAC等結果來評估樣品之抗氧化能力。實驗結果顯示,七種水萃物樣品中,總酚及總類黃酮含量最高均為白水木(花),分別為262.04±26.7 mg GAE/g extract及266.10±3.64 mg CE/g extract;在DPPH自由基清除能力最佳者為棋盤腳(花),其EC50為12.59±2.09 μg/mL;還原力效果最佳者為白水木(果實),其EC50為26.29±1.00 μg/mL;總抗氧化能力效果最佳為棋盤腳(花柄),TEAC值為1.70±0.02 mmol TE/g extract;螯合亞鐵離子能力測定後最佳為棋盤腳(花柄),EC50為0.85±0.06 mg/mL;而七種樣品在去氧核醣核酸保護上則都有效用,以白水木(果實)效果最佳。在抗菌試驗評估方面,發現棋盤腳(花柄)抑制革蘭氏陽性菌能力最強,抑菌圈大小約為13 mm。在抑制玻尿酸?上,顯示棋盤腳(花柄)和石栗(葉)之效果與沒藥醇能力相當。在細胞毒性試驗中棋盤腳(花柄)細胞存活率約為80%外,其餘樣品均無細胞毒性。
    This study mainly investigated the bioactivity of three species aqueous extracts which were collected from the tropical strand forest in Hengchun Peninsula, Taiwan, with hot water extracted approach. These species include Barringtonia asiatica L. Kurz (flower and pedicel), Tournefortia argentea L. f. (fruit, leaf, and flower), and Aleurites moluccana L. Willd. (leaf and flower).The antioxidant capacity of the aqueous extracts was determined by the readings of DPPH radical scavenging, ferrous ion chelating, and TEAC of each sample. The results showed that the Tournefortia argentea L. f. (flower) had the highest contents of total phenolic (262.04±26.7 mg GAE/g extract) and total flavonoids (266.10±3.64 mg CE/g extract) among all the seven tested aqueous extracted samples. The Barringtonia asiatica L. Kurz (flower) sample performed the best DPPH radical scavenging with EC50 at 12.59 ± 2.09 μg/mL. The Tournefortia argentea L. f. (fruit) sample had the most effective reducing activity, EC50 at 26.29 ± 1.00 μg/mL. The Barringtonia asiatica L. Kurz (pedicel) had the most powerful activity of total anti-oxidative capacity in 1.70 ± 0.02 mmol TE/g extract, also the most potent ability of ferrous ion chelating (EC50 at 0.85±0.06 mg/mL). All of the samples have certain degrees of capability on deoxyribonucleic acid (DNA) protection. The Tournefortia argentea L. f. (fruit) shows the best result than other tested samples. On the study of antimicrobial activity assay, the average zone of inhibition was completed by the paper disk diffusion approach. The Barringtonia asiatica L. Kurz (pedicel) reveals the strongest capacity against Gram-positive bacteria with size about 13 mm. On the inhibitory activity of hyaluronidase, the Barringtonia asiatica L. Kurz (pedicel) and the Aleurites moluccana L. Willd. (leaf) displayed the same effects as Bisabolol. For the cytotoxicity test, the results indicated that the Barringtonia asiatica L. Kurz (pedicel) had the cell viability with approximately 80%, as for the remaining samples were not cytotoxic.
