    Title: 牛樟芝化妝品原料開發之研究
    Development of Antrodia cinnamomea cosmetic formulation

    Abstract: 牛樟芝是台灣特有珍貴藥用真菌,含有大量的多醣體和三?類物質,如樟芝酸A、B、C、K (AntcinA、B、C、K) 等兩百多種生理活性成分。牛樟芝相關論文至2015年7月pubmed有292篇,台灣博碩士論文發表有374篇,其中對抑制發炎、抗氧化、抑制酪胺酸?等功效有許多的探討,這些機能都能應用在妝品原料的開發。本研究採用樟芝標準菌株BCRC35396,以合法取得之牛樟椴木,在潔淨廠房操作植菌程序。在木材前處理過程,可以取得牛樟木純露;牛樟芝培育過程可取得液態發酵菌種、牛樟芝菌膜、牛樟芝子實體等材料。為了評估運用在妝品的可能性,本研究進行樣本的化妝品安全性及功能性試驗,結果顯示在沙門氏菌安全性試驗皆無致突變性;以MTT法測定顯示樣品無細胞毒性。在功能性試驗以脂多糖 (Lipopolysaccharide, LPS) 剌激巨噬細胞RAW264.7持續24小時,測定細胞所生成的一氧化氮濃度,結果顯示牛樟芝子實體經由75% EtOH萃取,在受測濃度 (200μg/ml),一氧化氮濃度明顯下降;抗自由基試驗使用DPPH來評估受測材料的抗自由基能力,結果顯示受測樣本皆有良好的抗自由基能力。綜合評估,牛樟芝菌膜和牛樟芝子實體經75% EtOH萃取,具有成為優質化妝品原料的潛力。在面膜製作上添加子實體經75% EtOH萃取,試用滿意度調查上,使用後評價為很好佔44.17%、好佔29.58%;過敏性及斑點皮膚的受測人員使用後膚質有明顯改善。
    Antrodia cinnamomea is a Taiwan specialty medicinal fungus with more than two hundred bioactivities, such as polysaccharides, triterpenoids and Antcin A, B, C, K. Until April 2015, there were 292 and 374 in Pubmed and Taiwan national library of theses and dissertations, respectively. Most of studies focus on the anti-inflammatory, antioxidation and tyrosin kinase inhibition of A. cinnamomea. In this study, we attempted to investigate the potential application as cosmetic materials. A. cinnamomea strains (BCRC35396) was used in this study. A. cinnamomea were cultured on logs of Cinnamomum kanehirai after hydrosol extraction. Bioactivities were also obtained from liquid-state fermentation, the mycoderm and fruiting body of A. cinnamomea. To assess the potential application of those extracts as cosmetic materials, safety and functional tests were applied in following experiments. For safety test, results showed non-mutagenicity and non-cytotoxicity by MTT assay of Salmonella. The functional test, when used LPS to stimulate macrophages RAW264.7 to test the nitrogen monoxide concentration of cell for 24 hours. The result shows the fruiting body extracted by 75% ethanol (under the concentration of 200 μg/ml) significant decreased nitrogen monoxide concentration. Meanwhile, all samples showed good antioxidative activity in DPPH assay. The mycoderm of A. cinnamomea extracted by 75% ethanol also have the good potential anti-oxidant as cosmetic material. When mask manufacture contained the fruiting bodies (extracted by 75% ethanol), returned questionnaires showed there are divided into 41.17% and 29.58% to show well and good respectively. The participants with allergic and spots face have improved obviously in a month follow-up. In conclusion, for the best recovery function was achieved by using the fruiting body of A. cinnamomea (extracted by 75% ethanol) and on the other hand, for the best anti-oxidant function was obtained by extracting mycoderm from A. cinnamomea (by 75% ethanol).
