    摘要: 本研究係根據水溶性酸性煤焦色素 (water-soluble acid coal-tar dyes)之帶電性,利用聚丙醯胺膠柱(含5%單元體,pH6.7)做不連續電泳(Tris-glycine,pH8.0;10 ma/gel)以達到此類色素快速之分離與鑑定之目的。本法分離時間只需20~50分鐘,而色素含量在0.2?g以上即可檢出,同時又不受蔗糖、麥芽糖、弱酸、弱鹼、鹼性合成色素及天然色素等之影響,就所試的十四種合成色素中,有九種可完全分離,另外五種則可配合色帶之顏色及在酸鹼液中之變化而予確認,加上樣品處理簡單,實為食品及藥物中合成色素檢出之理想方法。本實驗同時發現利用丙酮處理可將結果長期保存。
    In this paper, fourteen authentic water-soluble acid coal-tar were studied by polyacrylamide gel disc electrophoresis (0101 M of Tris-glycine, ph8.0, containing 0.1% of NaCl; 10 ma/gel). It was found that 20-50 minutes were enough to separate most of them on one gel. And food additives were enough to separate most of them on one gel, and food additives or constituents such as sucrose (not more than 40%), maltose (not more than 20%), weak acid, weak alkali, natural dye or alkaline synthetic dye tested shown no or little effect.
    The other advantages of this method observed were easy performance, high reproducibility, small amount of dye being required, and simple sample preparation. Data indicated that polyacrylamide gel disc electrophoresis may be a suitable method to detect synthetic dyes in foods and drugs.
