    Title: 冬蟲夏草二次代謝物之生物活性研究

    Abstract: 近年來,冬蟲夏草被研究為傳統中藥或保健食品,其藥用價值已被多數專家肯定,然而許多研究也指出冬蟲夏草之菌絲體及子實體具有抗腫瘤、抗氧化、抗菌、免疫調節等功效。本論文研究目的為探討冬蟲夏草固態及液態培養的二次代謝產物,其有機相主要成分以及應用生物轉化的技術後生物活性之影響。於冬蟲夏草培養中添加六種中藥之水萃取物,以評估其生物轉化前後的生物活性。冬蟲夏草經由生物轉化二次代謝物9天的變化,在層析圖中,培養第7天開始持續至第9天,滯留時間26-33分鐘均有較多二次代謝物的吸收訊號,並針對培養第7天至第9天之生物活性試驗。中藥水萃物本身就具有良好的抗氧化能力,在DPPH活性結果顯示,固態培養成功的將冬蟲夏草生物轉化,其添加厚朴水萃物之清除率最佳為75%以上,其次為添加澤瀉、黃芩水萃物分別約為49%、47%之清除率。總酚含量測定的結果顯示,添加中藥水萃物之冬蟲夏草液態培養二次代謝物大部分在第7天的總酚含量最為顯著,分別為添加厚朴第7天約為197.2 mg/g與添加黃芩第7天約為170.7 mg/g,雖然代謝轉化成功的時間不一致,但經代謝轉化後總酚含量明顯提升。綜合上述實驗結果,我們認為冬蟲夏草之生物轉化代謝物具有深入探討的價值。
    In recent years, Cordyceps sinensis is used as traditional chinese medicine or health food. The medicinal value has been affirmed by many experts and researchers. However, many research results also pointed out that mycelium and sporocarp of C. sinensis were shown the following biological activities, such as anti-tumor, anti-oxidant, anti-virus and the immune regulation effect etc.The aim of this study is to explore the application of biotransformation and secondary metabolites of C. sinensis. The medium was added with water extract of six herbs to evaluate the biological activities of the biotransformation before and after. In the HPLC chromatogram, retention time between 26-33 minutes of culture from 7 to 9 days showed more absorption signals of secondary metabolites.The herbs extracts have good antioxidant capacity. The scavenging activity of DPPH of Magnolia officinalis solid medium is above 75%. Total phenolic content of herb-added liquid mediums at 7th day are the highest.
