    Title: 紅茄萃取物抗致突變及抗氧化能力分析
    Assessment of Antimutagenic and Antioxidation Activities of Salanum integrifolium Poir Extracts

    Abstract: 因紅茄的研究甚少且有越來越多人食用之趨勢,故本實驗利用紅茄水及50%、95%乙醇萃取物進行安氏試驗法(Ames test) 包含毒性、致突變和抗致突變試驗,並測定其抗氧化能力(總抗氧化能力、還原力、清除DPPH自由基能力和亞鐵離子螯合能力)與其抗氧化性成分(總多酚類和總類黃酮化合物)。實驗結果顯示在所選取的最高劑量範圍內,水萃取物、50%、 95%乙醇萃取物(5 mg/plate) 無論是否有添加S9 mix對Salmonella typhimurium TA98和TA100菌株不具有毒性也不具致突變性。在抗致突變方面,水及乙醇萃取物對直接型致突變劑(4-nitroquinoline-N-oxide)之抗致突變率分別 3.2~34.6% (TA98) 和4.1~37.5% (TA100)。但於添加S9 mix時水及乙醇萃取物對間接型致突變劑(2-aminoanthracene) 所誘發的致突變有明顯抑制作用,其抗致突變能力為10~89% (TA98) 和18.6~93.6% (TA100)。在抗氧化能力及其抗氧化成分分析實驗結果顯示,隨著紅茄萃取物濃度愈高,總抗氧化能力和還原力以及其DPPH清除自由基能力,以50%乙醇萃取效果最好,分別是總抗氧化力(3.937mM of trolox)、還原力斜率(1.412)、DPPH清除自由基能力 (101.31%),亞鐵離子螯合能力則以95%乙醇萃取效果最好。抗氧化性成分分析總多酚類含量則以水萃萃取物較高,其總多酚含量為(799.74 mg of GAE/g),總類黃酮含量則是以95%乙醇萃取效果最好,其總類黃酮化合物含量為102.97 (mg of rutin/g)。綜合以上所述,本試驗結果顯示無論在安氏試驗及抗氧化評估方面,紅茄萃取物都是隨著濃度越高越有抗致突變性與抗氧化能力,因此推測紅茄萃取物具有抗致突變的潛力且有良好之抗氧化能力。
    Salanum integrifolium Poir, solanaceae, solanum wild varieties, annual herb, origin to Africa、South America、Brazil, Taiwan was to Hualien. Uses include ornamental and medicinal, has anti-inflammatory analgesic, loose silt swelling effect, attending stomach pain, lymphadenopathy and other symptoms. The study was investigated the antioxidant and antimutagenic activity of Salanum integrifolium Poir water and ethanolic extract by several antioxidant activity methods and Ames test. Results indicate that microbial toxicity and mutagenicity assay (Ames test) showed that all the extracts(0.3125~5 mg/plate) of water, ( 0.3125~5 mg/plate) of 50% ethanolic, and (0.04~5 mg/plate ) of 95% ethanolic, with or without rat liver enzyme mixture (S9), had no toxicity and mutagenicity effect toward all tester strains (Salmonella typhimurium TA98 and TA100). Antimutagenic test of Salanum integrifolium Poir water extract showed inhibition effect toward TA98 against 4-NQO (the inhibition was 10.3~24.5%) and toward TA100 (the inhibition was 4.1~37.5%) against 4-NQO;Salanum integrifolium Poir 50% ethanolic extract showed inhibition effect toward TA98 against 4-NQO (the inhibition was 3.2~16.4%) and toward TA100 (the inhibition was 6.9~22%) against 4-NQO;95% ethanolic extract showed inhibition effect toward TA98 against 4-NQO (the inhibition was 9.6~34.6%) and toward TA100 (the inhibition was 9.1~17.7%) against 4-NQO. but water extract has lower exhibition effect toward TA98(the inhibition was 10~31%) against 2-aminoanthracene and toward TA100 (the inhibition was 18.6~44.6%) against 2-aminoanthracene;50% ethanolic extract showed the inhibition effect toward TA98 against 2-aminoanthracene (the inhibition was 14.3~65%) and toward TA100 (the inhibition was 29.9~69.6%) against 2-aminoanthracene;95% ethanolic extract showed the inhibition effect toward TA98 against 2-aminoanthracene (the inhibition was 33.7~89%) and toward TA100 (the inhibition was 44.6~93.6%) against 2-aminoanthracene. On antioxidant activity, results indicated that the 50% ethanolic and water extract showed a high total antioxidant capacity (779.07±0.03 and 799.74±0.01 mg of GAE/g) with total phenols method. And 95% ethanolic extract showed a high total antioxidant capacity with total flavonoid method. The TEAC of water and 50% ethanolic extract of Salanum integrifolium Poir, and values were 3.862 and 3.937 mM of trolox, respectively. Salanum integrifolium Poir results of DPPH scavenging capability: for 50% ethanolic extracts were the highest scavenging rate, with were 101.31%. Salanum integrifolium Poir results of Reducing power : for 50% ethanolic extracts were the high. 95% ethanolic extract of Salanum integrifolium Poir where showed effective chelating abilities on ferrous ions. In conclusion, mutagenic test showed the Salanum integrifolium Poir ethanolic and water extract had no mutagenicity effect toward all tester strains(Salmonella typhimurium TA98 and TA100). Results showed that ethanolic extract of Salanum integrifolium Poir had the strongest antioxidant capacity and antimutagenicity as well as and in a dose-dependent manner. At last, this study reveals that Salanum integrifolium Poir has the potential of relative functional-food industry because Salanum integrifolium Poir is safe and has antioxidative capability.
