摘要: | 本論文首先篩選血管收縮素轉化酶(Angiotensin Ⅰ-converting enzyme, ACE)抑制率的測定方法,比較乙醯乙酯、2,4,6-trinitrobenzenesulfonic acid (TNBS)和Pyridine與Benzene sulfonyl chloride (BSC)3種呈色法於測定發酵液之優缺點,以Pyridine與Benzene sulfonyl chloride (BSC)呈色法和HPLC測定法呈現良好的關係性。由159株植生型乳酸菌中篩選抑制ACE能力之菌株。在159株植生型乳酸菌中,血管收縮素轉化酶抑制能力達50 %以上的,有11株植生型乳酸菌;將11株植生型乳酸菌以12、16和20 mg/mL蛋白質濃度之培養基發酵,篩選出6株具有顯著抑制ACE能力之植生型乳酸菌,6株植生型乳酸菌B0014、B0015、B0059、B0060、B0096、B0137具有顯著性的ACE抑制能力。此外,將具有潛在性ACE抑制能力的不同素材,經過液化及糖化處理後,再以蛋白酶水解作為培養基,經此6株乳酸菌發酵後,選擇具有顯著性的ACE抑制能力的培養基。其中以綠豆、毛綠豆、黑豆和小米,將這5種素材製成培養基,以B0014、B0015、B0059、B0060、B0096、B0137植生型乳酸菌發酵後得到具有ACE抑制能力之發酵液。 Select the Angiotensin Ⅰ-converting enzyme (ACE) inhibition rate determination method, comparative acetyl ethyl, 2,4,6-trinitrobenzenesulfonic acid (TNBS) and pyridine with benzene sulfonyl chloride (BSC) 3 colorimetric method the determination of the advantages and disadvantages of fermentation broth, Pyridine and Benzene sulfonyl chloride (BSC) colorimetric method and HPLC assay showed a good relational. Investigate the Angiotensin Ⅰ-converting enzyme (ACE) inhibitory ability of 159 lactic acid bacteria (LAB) isolated from different plant resources. Among of 159 strains LAB, 11 strains showed the ACE inhibitory activity over 50%. Fermentation in different protein concentration medium, 6 strains from the above LAB had significant high ACE inhibitory ability. Strains of B0014, B0015, B0059, B0060, B0096 and B0137 showed high ACE inhibitory ability, and the R2 is 0.9644 between BSC-pyridine and HPLC method. Millet, black bean, mung bean and red bean have been widely used as major grains in Taiwan. In the present study, powder of grains was fermented with 6 strains of LAB after liquefaction and saccharification, and ACE inhibitory ability was measured. The results suggested that ACE inhibitory ability of millet and red bean could be increased by LAB. |
