    Abstract: 樹枝狀高分子的外圍官能基與空腔部位具有高的載藥能力,它們可以提高藥物在水中溶解度與生物可利用性,此外,樹枝狀高分子和藥物可以經由裝封、靜電相互作用、共價鍵結方式成為藥物輸送系統。聚胺基甲酸酯被廣泛應用於生物醫學各種植入的裝置,因為它具有生物可降解性與無毒性。本研究主要進行含不同世代及外圍官能基樹枝狀聚胺基甲酸酯的合成,利用傅立葉紅外線光譜(FT-IR)與核磁共振光譜儀(NMR)確定其化學結構。並進行以下之性質研究:酸鹼滴定分析緩衝能力、SEM粒徑分析、細胞毒性、藥物溶解度增加量、藥物負載及釋放動力。綜合以上研究資料,我們發現合成之樹枝狀聚胺基甲酸酯可以成為理想的藥物輸送系統。
    Dendrimers have high drug loading ability endowed by the large numbers of surface functionalities and interior cavities. They can render drugs greater water solubility and bioavailability. Furthermore, the dendrimers and drug molecules can including simple encapsulation, electrostatic interaction, and covalent conjugation, suitable as one of the drugs delivery systems. Polyurethanes have extensively been used in biomedical various implantable devices because they have biodegradable and nontoxic. The aim of this study is preparation of polyurethane dendrimers, the chemical structure of the dendrimer was indentified by fourier transform infrared spectroscopy (FT-IR) and nuclear magnetic resonance spectroscopy (NMR). We also studied various properties about the synthesized polyurethane dendrimers as acid-base titration to determine the buffering capacities, particle size, cytotoxicity, the increasement of drug solubility, drug loading and drug released kinetics. From above data, It is found that polyurethane dendrimers can be used as drug delivery system.
