摘要: | 本研究探討大花紫薇(Lagerstroemia speciosa)、木棉(Bombax ceiba Linn)、爪哇旃那(Cassia javanica L.)、阿勃勒(Cassia fistula Linn)、火焰樹(Spathodea campanulata)、火筒樹(Leea guineensis)與鳳凰木(Delonix regia)等七種天然植物,取其花朵經熱水萃取及冷凍乾燥後測定其生物活性之功效。研究目的是利用熱水萃取法評估這些植物花朵之水萃物是否同時具有抗氧化、抗發炎與抗菌之能力。實驗結果顯示,在抗氧化能力試驗方面,總酚含量以大花紫薇花(LS)之水萃物含量最多,為243.4±2.33 mg GAE/g sample,其次為火筒樹花(LG)之水萃物,為236.2±2.41 mg GAE/g sample;而總類黃酮含量以木棉花(BC)和火筒樹花(LG)之水萃物最多,分別是111.5±0.13 與99.74±0.41 mg CE/g sample;在清除DPPH自由基能力方面,以LS之水萃物之抗氧化能力最佳,EC50為14.60±1.59 μg/mL;在還原力試驗中,仍能以 LS 水萃物之還原能力最佳,EC50為12.52±0.04 μg/mL,其次為 LG水萃物,EC50為16.78±0.03 μg/mL;在總抗氧化能力試驗(TEAC)中,以LS之水萃物結果最佳,3.543±0.23 mmol TE/g sample;另外,在螯合亞鐵離子能力試驗,除了LS與LG之水萃物對於螯合二價金屬離子能力較好外,其餘樣品效果均不佳;而抑制脂質過氧化試驗,幾乎每個樣品的抑制百分率都有70%以上,只有火焰樹花(SC)之水萃物抑制率低於該數值以下。在抗發炎能力方面,顯示大花紫薇花(LS)、木棉花(BC)、火筒樹花(LG)與鳳凰木花(DR)之水萃物對玻尿酸酶均有明顯的抑制效果,亦即此四者均具有很好之抗發炎能力。而在抗菌試驗方面,以紙錠擴散法評估LS之水萃物,發現其對革蘭氏陽性菌與陰性菌之抑菌能力最強,抑菌圈大小分別約為13.67 mm與18.67 mm,其餘樣品的抑菌圈都在9~11 mm之間;而最小抑菌濃度(MIC)與最小殺菌濃度(MBC)之試驗,均大於1 mg/mL。最後,在細胞毒性試驗,七種植物花朵水萃物對於細胞存活率,除了爪哇旃那花(CJ)與阿勃勒花(CF)之水萃物對細胞有低毒性,其存活率為60至80%,其他均大於80%以上。綜合以上實驗結果得知,在所有樣品之中,以大花紫薇花(LS)與火筒樹花(LG)的水萃物質對於抗氧化、抗發炎與抗菌能力均為最佳。 This study aimed to screen for the biological activities of anti-oxidation, anti-inflammation and anti-microorganism from Lagerstroemia speciosa (LS), Bombax ceiba Linn (BC), Cassia javanica L. (CJ), Cassia fistula Linn (CF), Spathodea campanulata (SC), Leea guineensis (LG) and Delonix regia (DR). Then, we use the method of hot water extraction to get the crude extracts of each sample’s flowers.For the results of anti-oxidations, the total phenolic contents of LS (243.4±2.33 mg GAE/g sample) and LG (236.2±2.41 mg GAE/g sample) were higher than other samples. In addition, the result of the total flavonoid contents displayed that the extract of BC and LG had more flavonoids in 111.5±0.13 and 99.74±0.41mg CE/g sample, respectively. These results showed that LS had the best biological activities of DPPH radical scavenging in all the test samples, the EC50 at 14.60±1.59 μg/mL. On the reducing activity test, the best value of the EC50 was in 12.52±0.04 μg/mL and 16.78±0.03 μg/mL for LS and LG, respectively. The LS had the best activity of total anti-oxidative capacity in 3.543±0.23 mmol TE/g sample. For the ferrous ion-chelating ability assay, the aqueous extracts of LS and LG were shown in obvious effect of metal ion chelating, while other samples are lower than this result. Almost the extracts, for activity of anti-lipid peroxidation test, had more than the inhibition rate of 70%, but the water extracts of SC is lower than this value.On the study of anti-inflammation, our results displayed that the water extracts of LS, BC, LG and DR owned a good ability to inhibit the activity of hyaluronidase. Experimental study on antimicrobial activity assay, the average zone of inhibition was produced by the paper disc diffusion method, the aqueous extract of LS revealed the strongest capacity against the growth of Gram-positive and Gram-negative bacteria, with a clear zone of 13.67 mm and 18.67 mm, respectively. However, the inhibitory zones of other samples were between 9 and 11mm. For the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) assay, the concentration of all the extracts were greater than 1mg/mL. Finally, for the cytotoxicity test, our results showed that the cell viability were above 80% in all samples. However, CJ and CF indicated low toxicity for cells with the survival rate of 60 to 80%. According to the above results, we find that LS and LG are the best antioxidants, anti-inflammatory and antibacterial agents among the test samples, respectively. |
