    標題: 消化藥劑中澱粉酶分析方法之探討

    摘要: 本報告乃嘗試用澱粉-碘反應以分析原料或製劑內澱粉酶活性,發現較一般藥典所敘述的方法為便捷,同時亦有定量的效果,主要的結果包括
    (1) Diastase或pancreatin只要在1 μg以上即可在室溫於20分鐘內利用本法檢出其含量。
    (2) 超音波處理可應用於腸衣錠內酵素之抽取。
    (3) 含澱粉酶之酵素液,於室溫中放置,其活性會逐漸下降,若於冰箱內保存則至少可維持兩天以上。
    (4) 當不含制酸劑的消化藥用擬胃劑(USP;不含胃蛋白酶)抽取時,其所含之澱粉酶活性會消失。
    (5) 消化劑間含有不等量的澱粉酶活性,大部分經本法分析後其澱粉酶含量均較標示量為低。
    In this study, starch-I reaction is demostrated as a better method in quantitative analysis of amylase activity in both raw material and digestant than the methods presented in several pharmacopoeia. By means of this more convenient procedure, the amylolytic abilities of several popular digestants are analyzed. Other significant findings as followed are also discussed.
    (1) Diastase or pancreatin as low as 1 fug can be detected within 20 min at room temperature.
    (2) Sonification is demostrated to be a suitable procedure for preparation of enteric coat homogenate.
    (3) On standing at room temperature, amylolytic activity of pancreatin solution decreases gradually. When the digestant without antacids ( such as NaHCO3 ) is extracted with gastric fluid (USP ;without pepsin ), all :he amylolytic activity is lost.
    (4) The digestants tested show different significantly in amylolytic activity, and some of them preserve an activity less than 50% of enzyme amount labeled.
