    摘要: 近年來國人視溫泉具養生保健概念而形成一股泡湯熱潮,溫泉業者為了能迎合國人溫泉浴的消費習慣,偏重多樣化溫泉浴池及自然情境設計,並強調原泉直接供給消費使用的經營型態。然而,忽略了抗菌材質及衛生管理的重要性,導致病菌感染事件,時有所聞。本研究擬從中現今業者和消費者常忽略的溫泉衛生管理及消費者嗜好原泉浴的習慣中,同時折衷參考台灣溫泉浴池經營和水質衛生管理現況,探討溫泉水質條件對生物膜生成影響概念。
    研究採EHEC O4大腸桿菌為水質污染菌種,將EHEC O4培養於分別添加3 ml的NaCl溶液、北投溫泉、泰安溫泉、關子嶺溫泉、寶來溫泉、四重溪溫泉、知本溫泉及安通溫泉之12孔培養皿孔內,並於第3天、7天檢測其在12孔培養皿上所生成的生物膜(crystal violet染色法),以評估溫泉對細菌生長之影響。本研究結果發現,NaCl溶液培養之生物膜生長情形在第1天開始呈線性增長關係。進一步比較NaCl溶液和各種不同溫泉泉質對細菌生物膜生長之影響評估,在第3天的染色後吸光值數值北投(0.17)、泰安(0.27)、關子嶺(0.27)、寶來(0.28)、四重溪(0.25)、知本溫泉(0.29)及安通(0.44),比對照組NaCl溶液(0.63)皆呈現抑制作用,但在第7天時,泰安(1.66)、寶來(1.04)及四重溪(0.92)溫泉染色後吸光值相較對照組(0.78)數值較高,而北投(0.18)、關子嶺(0.36)及知本(0.48)溫泉則仍呈現抑制作用,安通 (0.75)溫泉則與對照組相近。綜合本研究結果顯示,台灣大部份溫泉區之溫泉具有抑制細菌生物膜生長作用,但也有少數溫泉具有促進生物膜生長作用,其詳細之作用機制則待更多研究證實。
    In recent years, citizens in Taiwan embrace the idea of hot-spring for health care and develop the habit of spring-spa consumption. The spring industries emphasize on providing diversified designs of spa hot tub and natural environment scenario to attract Taiwan’s consumers, and yet, neglect the importance of antibacterial material usage as well as hygiene management. Such business ignorance leads to the frequent incidence of pathogen infection to spring consumers. This research discuss concept of the spring water conditions that affect the bio-film forming in an attempt to put forth a good managerial approach between the spring tub design and water sanitation by views of spring hygiene management, Taiwan consumers’ consuming habits of original spring, current spring-industries’ operation model and water sanitary management.
    In this study, the E. coli strains (EHEC O4) was used an indicator of water pollution. The EHEC O4 was added in the 12 wells culture dish hole, that each was included 3 ml of NaCl solution (control group) and others hot spring group (Beitou, Taian, Guanziling, Baolai, Sichongxi, Zhiben, Antong). At the 3 days, 7 days, to detect the generated biofilm in the petri dishes of 12 holes (crystal violet, staining) to assess the impact of hot springs on the growth of bacteria. The results of this study found that the linear growth relationship of NaCl solution culture biofilm growth situation in one day. Further compare the NaCl solution and a variety of spring quality impact assessment of bacterial biofilm growth, absorbance values Beitou (0.17) in the first three days after dyeing, Taian (0.27), Guanziling (0.27), Baolai (0.28 ), Sichongxi (0.25), Zhiben (0.29) and Antong (0.44) than the control group, NaCl solution (0.63) are showing inhibition, but in seven days, Taian (1.66), Baolai (1.04) and Sichongxi (0.92) absorbance in the Hot Springs stained compared to the control group (0.78) higher values, while the hot springs of Beitou (0.18), Guanziling (0.36), Zhiben (0.48) is still showing inhibition, Antong (0.75) hot springs similar to the control group.
    The results shown that most of the hot spring area in Taiwan played the role to inhibit bacterial bio-film growth, but there are a few hot springs promote the role of bio-film growth. Furthermore, the detailed mechanism of action is to be more research.
