    標題: 白藜蘆醇對於小鼠維生素E營養狀況和抗氧化系統的影響

    摘要: 維生素E又稱生育醇(Tocopherol),為人體必需的脂溶性維生素,也是體內最重要的抗氧化劑,可減輕體內自由基對人體造成的傷害,降低血液中的脂質過氧化情形。有研究指出許多酚類化合物能夠提生動物體內維生素E的營養狀況;白藜蘆醇也為多種多酚類化合物的其中之一。白藜蘆醇(Resveratrol)產生於葡萄葉表皮和漿果果皮中,除了具有抗氧化能力之外,亦有研究指出白藜蘆醇具有抗肥胖的作用。但是在白藜蘆醇相關的論文中有關維生素E代謝的研究很少;目的:因此本實驗室欲探討白藜蘆醇對小鼠體內維生素E營養與抗氧化系統的影響。方法: 將24隻7週大雄性C57BL/6J mice 適應兩週後依飼料不同分為下列三組: LF組 ( 5% Fat )、 HF組 ( 20% Fat )和 HF-R組(20% Fat,0.04% Resveratrol)。飼養期八週,於最後一週收集尿液,犧牲當天取得血液、肝臟、脂肪及其他組織。分析項目如下:維生素E的營養狀態 ( 各組織的維生素E濃度、肝臟及血液中的TG、TC等 )、抗氧化系統的影響 ( 如:肝臟的抗氧化酵素活性、TBARS、尿液8-isoprotane等 )及血漿adiponectin和肝臟α-生育醇轉移蛋白(α-TTP)、細胞色素P-450 3A11 (CYP 3A11)、glutathione-S-Transferase 。結果: HF-R組的心臟、脾臟生育醇濃度顯著低於其餘兩組( P<0.05 ),GSH濃度亦呈現下降的趨勢,然而體內組織TBARS含量並無顯著差異;肝臟的抗氧化酵素CAT活性雖以HF-R組有較高的趨勢,但與其餘兩組無顯著差異。HF組的adiponectin濃度顯著低於LF組,但是HF-R組卻可以提升adiponectin的濃度。在肝臟α-TTP蛋白質含量結果顯示,三組之間並無顯著差異。而在肝臟CYP3A11蛋白質含量結果顯示,雖然在數值上依序以HF-R組較高HF組為次,但統計上並無顯著差異。另外在COX-2的心臟和腎臟在數據上都是以HF組來的較高而HF-R組為次,不過亦無顯著差異;肝臟的COX-2蛋白質量三組間亦無顯著性差異。結論: 從本實驗結果指出,在高脂飲食中添加白藜蘆醇會降低小鼠體內維生素E濃度,但是並未見有脂質過氧化物質增加之情形,表示白藜蘆醇雖未降低體內的脂質過氧化情形但是確實會影響體內維生素E營養。白藜蘆醇在維生素E代謝相關蛋白質α-TTP、 CYP3A11的調節或吸收利用並無明顯影響,至於白藜蘆醇是如何調節體內維生素E營養狀況則仍須進一步探討。
    Vitamin E, also known as tocopherol, is an essential lipid-soluble vitamin and is the most important antioxidant in the body. Several studies revealed that polyphenol showed a sparing effect of vitamin E in animals. Resveratrol ,one of various polyphenols, is being produced in the grape leaf’s epidermis and the berry fruit peel. Resveratrol was reported to having an anti-oxidize ability, also anti-obese ability. The effects of resveratrol feeding on the vitamin E status in animals has not been examined. The purpose of this study was to investigate the effect of resveratrol on the vitamin E status and antioxidant systems in mice. Twenty-four seven-week-old male C57BL/6J mice were assigned to three groups: Low-Fat group (LF, 5 % Fat), High-Fat group (HF, 20% Fat), Resveratrol group (HF-R, containing 20% Fat with 0.04% Resveratrol). The animals were fed with experimental diets for 8 wk. The results showed that α-tocopherol concentrations of heart and spleen in HF-R group were significantly lower than the other two groups (P <0.05). TBARS concentrations of the tissues were no significant differences among the three groups. In HF-R group, the activity of antioxidant enzymes, CAT, in the liver showed higher than the other two groups (p>0.05). The adiponectin levels in the HF group showed significantly lower than that in LF group,however, the adiponectin levels in the HF-R restored and as high as the LF group. The protein levels of liver α-TTP and CYP3A11 showed no significant differences among the three groups. There is no significant difference in the COX-2 protein in liver, heart and kidney among the three groups. In conclusion, supplementation of 0.04% resveratrol in high-fat diet reduced the vitamin E concentrations in heart and spleen but no effect on increasing lipid peroxidation. Resveratrol showed no influence on the vitamin E metabolism-related protein such as α-TTP、CYP3A11. Therefore, how the vitamin E status was regulated by resveratrol needs further investigation.
