摘要: | 丹參學名Salvia miltiorrhiza Bunge,為脣形科鼠尾草屬植物,以乾燥根莖部入藥。丹參在中草藥應用最早記載於《神農本草經》,載明其功效去瘀止痛、活血通經、清心除煩。本研究丹參取材國內中藥廠自大陸進口及台灣花蓮地區種植之丹參。
台灣及大陸丹參以不同比例之溶劑( 95 %酒精、50 %酒精、純水)萃取。定性分析此兩種丹參之紫外線吸收圖譜(200 nm ~ 700 nm)波形相近。進一步以薄層層析法(Thin Layer Chrom atography,TLC)鑑別結果顯示此兩種來源之丹參皆含有丹參酮IIA(Tanshinone IIA)與丹酚酸B(Salvianolic Acid B)之指標成分,由此兩種鑑別方式可判定所取得之植物皆為丹參。
定量分析使用高效液相層析儀(High Pressure Liquid Chromatography,HPLC)得丹參酮IIA含量與萃取溶劑有關,尤以台灣丹參95 %酒精萃取物含量最高(10.49 mg/g);台灣丹參丹酚酸B含量優於大陸丹參(TDD1-121.82 mg/g、CDD1- 101.07mg/g);總酚酸含量以台灣丹參其相對沒食子酸含量最高(GAE值:73.52 mg/g)。
台灣丹參酒精萃取物清除ABTS陽離子自由基以及清除DPPH自由基能力之SC50分別為0.197 mg/ml、SC50 0.094 mg/ml為各丹參萃取物中抗氧化活性較佳。在抑制酪胺酸酶試驗台灣丹參萃取物濃度0.5 mg/ml時,各丹參萃取物抑制率可達66%以上。抑菌實驗,台灣丹參及大陸丹參95% 酒精萃物濃度為10 mg/ml時對痤瘡桿菌有具抑菌作用 (TDD1 抑菌圈11.3 mm、CDD1抑菌圈11.7 mm)。安全性評估,各丹參萃取物濃度0.5 mg/ml時對人類纖維母細胞WS1以及CCD-966SK細胞存活率皆有53%以及67%以上,顯示丹參是安全性高之植物。
本土台灣乾燥丹參萃取物應用於配方中,對配方並進行功效性評估。將台灣乾燥丹參不同溶劑萃取物進行保養乳液配方調製,並測試四週後其皮膚功效評估(黑色素、紅色素、表皮水分含量及皮膚彈性)有改善,且使用期間並無不良反應,長期使用添加丹參之保養乳液其低刺激性故可開發應用在保養化粧品。 Dan-shen, the dried root of Salvia miltiorrhiza (Fam. Labiatae),used in traditional Chinese medicinal herbs. The traditional curative functions were invigorate blood , remove stagnation and clear heat in the heart. In this study, the roots of Dan-shen was imported form China and planted in Hualien,Taiwan.
The roots of Dan-shen were extracted with three solvents (95 % ethanol, 50 % ethanol and water) .The Dan-shen was qualitqatively analyzed with UV absortion and thin layer chromatography (TLC).The UV waveform( 200~700 nm) of Taiwan and China Dan-shen extracts were similar. In TLC, two kinds of the Dan-shen had Tanshinone IIA and Salvianolic Acid B. Those Dan-shen from different area were indeed Dan-shen.
Dan-shen extracts were quantitatively analyzed with high performance liquid chromatographic (HPLC). The tanshinone IIA of Taiwan Dan-shen 95% ethonal extract (TDD1) contained 4.94 mg/g.Salvianolic Acid B in Taiwan Dan-Shen was more than China Dan-Shen.(TDD1-121.82 mg/g,CDD1-86.05 mg/g). In total phenol content assay, the extractions of Taiwan Dan-shen 95% ethanol extracts showed the highest-level value 73.52 mg of GAE/g.
Nine of Dan-shen extracts compared the antioxidant activity, tyrosinase inhibition activity, antibacterial activity and cell survival. The ABTS radical scavenging ability and DPPH radical scavenging capacity showed the SC50 of TDD1 lower than others ( SC50 0.197 mg/ml and SC50 0.094 mg/ml). Inhibition tyrosinase assay showed the inhibition percentage of the Taiwan Dry Dan-shen extracts were over 66 0 % at 0.5 mg/ml. Antibacterial activity:both of Taiwan and China Dan-shen 95% ethanol extracts exhibited Propionibacterium acne at 10 mg/ml (TDD1:11.3 mm,CDD1: 11.7 mm). In MTT assay for cells survival (WS1 and CCD-966SK), these Dan-shen extracts did not cause cellular dieth when dose at the 0.5 mg/mL, the cells survival rates over 53.54% and 67.2%,showed they are safety for cell.
Based on all results, the Taiwan Dry Dan-shen had higher activity than others. The study selected Taiwan Dry Dan-shen extracts made skincare lotions and tested human efficiency evaluation.After using 4 weeks, the skincare lotions with Taiwan Dry Dan-shen extracts had improvement effect on Melanin, Erythem, Corneometer value and Skin Elasticity.The volunteers used these skincare lotions with Dan-shen extracts had no irritating ,showed the product was safety for the cosmetic purposes. |
