    Title: 固態培養白樺菇之抗發炎、免疫調節與抗糖尿病活性及其相關分子訊息傳遞途徑之研究

    Abstract: 白樺菇(Inonotus obliquus)是一種名貴的藥用菇類,也為開發保健食品的一重要材料。民間一般認為白樺菇具有抗癌、免疫調節及抗糖尿病等功效。有關白樺菇之研究,雖目前發表之SCI 論文已有七十幾篇,然而大部分的研究均著重在液態培養白樺菇之活性,固態培養白樺菇之抗發炎、免疫調節及抗糖尿病活性及其相關分子訊息傳遞途徑尚不清楚,因此值得深入研究。本研究目的為(1)利用LPS-誘導人類單核球(THP-1)細胞探討固態培養白樺菇多醣體(SIOP)之抗發炎活性及其分子作用機制,含MyD88、 MAPK、NF-κB 與STAT-1 等途徑之訊息傳遞反應;(2)以動物實驗模式探討SIOP 之非特異性免疫與特異性免疫反應;及(3)以第一型糖尿病細胞模式探討SIOP 對STZ-誘導的大鼠胰島瘤(RIN-m5F)細胞之抗氧化壓力及細胞凋亡分子機制,與以第二型糖尿病細胞模式探討SIOP 對小鼠前趨脂肪 (3T3-L1)細胞之葡萄糖吸收及胰島素接受器分子訊息傳遞途徑之影響。為進一步研究固態培養白樺菇之藥理功能,希望國科會能再繼續提供研究經費,以便有系統的探討SIOP 之抗發炎、免疫調節與抗糖尿病活性,以及其相關分子訊息傳遞途徑。此研究結果將有助於固態培養白樺菇成為高經濟價值保健食品之重要資料依據。
    Inonotus obliquus, a high value medicinal fungus, is an important ingredient for health food development. It is generally believed to possess anti-cancer, immunomodulatory, anti-diabetes and other therapeutic properties. Regarding research on I. obliquus, although currently over 70 SCI articles have been reported on its bioactivities, many of these studies focused on biological activities of liquid cultured I. obliquus, the molecular signal transduction pathways of the anti-inflammatory, immunomodulatory and anti-diabetic activities of solid cultured I. obliquus remain unclear. Therefore, an insight study in these properties is needed. The aims of this study comprise:(1) to examine the anti-inflammatory activities of solid cultured I. obliquus polysaccharides (SIOP) and their molecular signaling cascades, such as MyD88, MAPKs, NF-κB and STAT-1 pathways in the LPS-induced human monocytic cells (THP-1); (2) to examine the effects of SIOP on the innate and adaptive immune responses in animals; and (3) the examine the anti-type I diabetic effects of SIOP on the oxidative stress and cell apoptotic mechanisms in the STZ-induced RIN-m5F cells, and the anti-type II diabetic effects of SIOP on the glucose absorption and the molecular transduction pathway of insulin receptor in 3T3-L1 cells. In order to continue our research works on pharmacological properties of solid cultured I. obliquus, we therefore hope that NSC would further provide financial support for our studies on the anti-inflammatory, immunomodulatory and anti-diabetic activities of SIOP and their related signaling transduction pathways. Results obtained from this study will provide vital information for the development of solid cultured I. obliquus into high economic value health food products.
