    Abstract: 中藥方劑由於組成成分複雜,劑型﹑質量標準上未能符合國際市場的要求;迄今仍未進入國際醫藥主流市場,因此若能如西藥水準,對每一藥物均能了解其作用機轉、有效成分、使用計量、副作用、交互作用之後,就能加快中藥現代化進程。近年來,因基因體及蛋白質體學的蓬勃發展,資料庫越來越完備及質譜分析技術的提升,中草藥以及天然物等小分子可以利用不同的質譜模式做精確且高靈敏度的定性定量分析;質譜技術更可被大量應用於分析生物分子;使得在短時間內進行大規模蛋白質功能探討不再遙不可及。因此本研究將以現代蛋白體質譜技術(如二維電泳、In gel digestion及液相層析質譜技術等)進行中藥方劑化學成分的研究;利用耐輻射奇異球菌的二次生長現象做為模型,探討中藥方劑對其生長所調控的代謝途徑及藥理機制之驗證,再根據實驗數據及方劑療效選擇適合的某種病症,建立與某種病症相對應的量化藥理模型,確定一種或多種藥效觀測指標及其活性評估標準,最終闡明中醫藥治療疾病的配伍規則和作用機制,建立中草藥研究資料庫,為中醫現代化提供理論和技術平台。
    The rapid growth of proteomics field provides an array of new tools for the integration of traditional Chinese medicine (TCM) with modern technology and systems biology, and is potentially advancing the progress of modernization and internationalization of TCM. This study has applied 2DE-based and bottom-up proteomics in the phenomenon of TCM-induced cell division on Deinococcus radiodurans, including the facilitation of herb authentication and identification, the exploration of pharmacological mechanisms of the actions of TCM, and the profiling of protein expression following TCM treatment in vitro. The research effort here presents a better insight to the integration of TCM and western medicine and develops a better way to study the molecular basis of syndrome.
