    標題: 植生型乳酸菌粉末化之研究及其功能性評估

    摘要: 本研究探討乳酸菌液態發酵的培養基、發酵條件及影響菌落數之乾燥條件及賦形劑,並評估貯存過程中,菌粉的活菌數及耐酸耐膽塩等特性。配方A (15% 脫脂乳粉、15% 海藻糖、15%乳糖)及配方B (20%脫脂乳粉、20% 果寡糖)複合賦形劑配方添加於B0007菌株,經冷凍乾燥後,B0007在賦形劑配方A、B的試驗,活菌數分別為2.65×1011及4.2×1011 CFU/g,而回收率為1.4至1.5%。而採用新型專利(M300796)之冷熱合一噴霧造粒乾燥機來製造乳酸菌粉,使用賦形劑配方D:(脫脂乳粉40%+玉米澱粉40%+果寡糖20%)進行乾燥,B0007菌數可達到3.25×1010 CFU/g,其回收率為35%;B0022利用配方H:(脫脂乳粉45%+麥芽糊精45%+果寡糖10%)進行乾燥期菌數量可達到4.25×1010利用兩種乾燥方式處理後進行貯存性試驗並定期取樣分析菌數及功能性評估,發現B0007或B0022添加配方A及配方B賦形劑進行凍乾處理,貯存於4℃,B0007菌粉、B0022菌粉貯存4個月菌數可達到1×107CFU/g 以上,研究發現冷凍乾燥B0007、B0022菌粉在貯存期間,皆具有耐酸及耐膽塩以及膽固醇吸附能力。噴霧乾燥部分,發現B0007菌株在賦形劑配方D的保護之下,於4℃環境下貯存到第6個月後的菌數可達到3.65×108 CFU/g,添加配方H之B0022菌株貯存到第6個月菌數可達到1.2×107 CFU/g。噴霧乾燥貯存6個月後的菌粉進行耐酸耐膽塩試驗,B0007及B0022菌數也可達到1×108 CFU/g以上,且進行膽固醇吸附試驗,研究結果可得知噴霧乾燥後的菌粉皆具有膽固醇吸附能力;由此可以得知本試驗兩株菌株在經過噴霧乾燥及冷凍乾燥處理後之菌粉,經過人體腸胃道的胃酸及膽塩環境下皆可以存活下來且具有吸附膽固醇之能力。
    The purposes of this study were to investigate the effects of medium compositions, fermentation conditions, different excipients and dry methods to the survival of lactic acid bacteria and to evaluate the viable counts, acid and bile tolerance during storage test by different dry conditions and excipients.
    Excipient A group (15% skim milk, 15% trehalose and 15% lactose) or B group (20% skim milk powder and 20% oligosaccharides) was added into the fermented culture and then freeze-dried. The cell viable counts and yield of cells were determined after freeze-dried. The results showed that the cell viable counts of B0007 with excipient A group and B group were 2.65×1011 and 4.2×1011 CFU/g and yield of cells were 1.4% and 1.5%, respectively . The powder of B0007 was also prepared by spray-dried method, the cell viable counts, 3.25×1010 CFU/g, and the yield of cells 35%. The powder of B0022 was also prepared by spray-dried method, the cell viable counts, 4.25×1010 CFU/g. In storage test, cell viable counts of B0007 and B0022 with A group or B group were over 1×107 CFU/g with freeze-dried after incubation of 4 months. For spray-dried powder after incubation of 6 months at 4℃,the cell viable counts of B0007 (with excipient D group: 40% skim milk, 40% corn starch, 20% oligosaccharides) and B0022 (with excipient H group 45% Skim milk 45% Maltodextrin 10% Oligosaccharides) were 3.65×108 CFU/g and 1.2×107 CFU/g. From the acid tolerance test, bile tolerance test and cholesterol absorption test, all the sample powder B0007 and B0022 with different excipients or different dry method showed that B0007 and B0022 are acid tolerance, bile tolerance and cholesterol absorption after storage.
