    標題: 天然物萃取及多酚類化合物應用於預防雄性禿保養品之細胞與動物模式評估
    Evaluation of Natural Product Extracts and Polyphenol Compounds on Prevention of Androgenetic Alopecia by Cell Culture and Animal Model

    摘要: 雄性禿(Androgenetic alopecia,AGA)為最常見的掉髮原因,佔男性禿頭的95%以上,經許多研究發現造成雄性禿主要的元兇是5α-還原酶 (5α-reductase)催化睪固酮 (testosterone,T)形成更具活性的二氫氧睪固酮 (dihydrotestosterone,DHT),而具活性的DHT會與真皮毛乳頭上的雄性激素接受體(androgen receptor)結合導致毛囊微小化,造成雄性禿症狀。
    而5α-reductase依其特性可分為Type I和Type II,此次實驗主要是利用本實驗室於2006年由涵潁建立5α R2的穩性細胞株及於2008年由雨儒建立5α R1的穩性細胞株作為測試抑制5α-reductase的實驗平台,與2009年由錦婷挑選出適當的轉換率分析平台,以作為迅速篩選多種中草藥萃取物和合成物能有效抑制5α-reductase的工具。
    另外,首先需將各萃取物及合成物需由MTS分析挑選出細胞存活率約80%以上的濃度來作為抑制5αR1及5αR2成分篩選的起始濃度。共篩選了九種天然物及三個多酚類化合物,結果為本省原生種萃取物(A)在濃度為1.10 mg/ml時對於5αR2有IC50的抑制效果。再更進一步選擇七週大的母鼠C57BL/6J mice進行動物實驗,結果顯示天然萃取物(A)對於小鼠背部毛髮有促進毛髮生長的效果。
    Androgenetic alopecia (AGA) is the most common causes of hair loss, accounting for more than 95% of male baldness. Many studies have been reported that the main reason causing male bald is the 5α-reductase, which catalyzes the conversion of testosterone (T) into more active dihydrotestosterone (DHT). Active DHT binds to the androgen receptor on dermal papilla, then leads to hair follicle miniaturization and subsequent Androgenetic alopecia.
    According the characteristics of 5α-reductase, they are divided into Type I and Type II. In this study, we used previously established 5α-reductase enzyme conversion assay and human embryonic kidney 293 cell lines stable expressing either Type I (pflag-CMV2-5αR1) or Type II (pflag-CMV2-5αR2) 5α-reductase, to quickly screen the potential inhibitors of 5α-reductase in several polyphenol compounds and extracts from several Chinese traditional herbs.
    First, using MTS assay to evaluate the cytotoxic effects of sample. Then, screening nine kinds of the natural products and three polyphenol compounds, our experimental results showed that the IC50 of enzyme only the extracts from nature product extract (A) contain inhibitor activities for 5αR2 at the concentration of 1.10 mg/ml. Finally, candidates will be then tested in seven-week-old female C57BL/6J mice. The results showed that nature product extract (A) proven ability to promote hair regrowth.
