    標題: 利用雞毛分解菌行生物分解作用生產胺基酸之最適條件探討

    摘要: 本研究系利用本實驗事先前自土壤終篩選出之雞毛分解菌:AS-B4,BS-B1,CS-B2及CS-B4四株菌,進一步對其分解雞毛產生胺基酸之最適操作條件作一個探討。所探討之操作條件包括:溫度、pH、菌種儲存穩定性、雞毛濃度及培養天數等項目,並依所得最適條件之組合來建立一個簡易生物反應槽,期能提高雞毛分解效率及胺基酸之產量。由最適溫度試驗結果得知:AS-B4,CS-B2及CS-B4均以37℃時之雞毛分解效率最高,而BS-B1則以25~37℃最高;並發現雖同樣在最適溫度下,CS-B2、BS-B1、CS-B2及CS-B4之雞毛分解效率均明顯較AS-B4為高。於initial pH試驗中,發現BS-BI,CS-B2及CS-B4均以pH8.5左右最適;其中CS-B2之pH適應性最廣( pH7.1-8.5 )。而菌種穩定性試驗結果則顯示CS-B2之貯存穩定性最高。綜合以上結果,後續試驗選用適於常溫操作之BS-BI及雞毛分解效率和穩定性均較高之CS-B2,以利經齊需求。於雞毛濃度及培養天數試驗中顯示: Bs-BbI及cs-B2以2.5%之雞毛濃度培養老天穀合乎經濟效益。最後設計並進行簡易生物反應槽之操作,結果發現: CS-B2之雞毛分解效率可提高至78.7% ; BS-B1則為76.0% ,頗具開發潛力,值得進一步研究。
    The purpose)se of this research is to set up the optimum conditions for production of amino acids by biodegradation with four previous selected feather-degrading bacteria (AS-B4, BS-B1,CS-B2 and CS-B4). The operating conditions included temperature, pH, storage stability, concentration of feather and incubation time. In the temperature test, the results showed that theoptimum temperatures of AS-B4, CS-B2 and CS-B4 were 37。C; BS-B1 was 25-37。C. Because biodegradation efficiency of AS-B4 was the lowest, we deleted AS-B4 in the later tests. In initial pH test, the results indicated that the optimum pHs of BS-B1, CS-B2 and CS-B4 were pH8.5. In storage stability of feather-degrading bacteria test, the result showed that the stability of CS-B2 was the highest. Thus, both BS-B1 and CS-B2 were selected for the later tests to achieve the economical requirement. The concentration of feather and incubation time test showed that 2.5% of feather powder and 9 days of incubation time was the most economical conditions. Finally, the biodegradation eMciency of CS-B2 and BS-B1 was up to 78.7% and 76.O%, respectively, with a simple bioreactor.
