本研究利用分光光度計嘗試建立一套快速、簡便之抗氧化活性之測定方法。本法基本原理為測定抑制亞麻油酸之過氧化反應之能力。將亞麻油酸、span 60、抗氧化劑之混合物經真空乾燥以形成薄膜,於60℃下加熱90分鐘後,利用分光光度計測量234mm之吸光值,換算為亞麻油酸過氧化物之產量,抗氧化活性則由抑制亞麻油酸過氧化物產量之比率加以計算。應用本法分析BHA、BHT及維生素E等抗氧化劑之抗氧化活性發現與其濃度成正相關。因此本法可應用於快速檢定油溶性抗氧化物之抗氧化活性。 A rapid photometrlc assay for evaluatlng antloxldatlve actlvity was develop. This method was establlshed prlnclpally on the inhlbltlon capacity in the pemxidation of linolec acld. A mlxture of linoleic acld and span 60 was vacuum dried to form a thin film in a test tube. The film mixture was then heated at 60 C for 90 min. with a heating block. The linoleic acld peroxide formed was measured with spectrophotmeter at 234 mm. Antioxldative acilvity of BHA, BHT and dl-a-tocopherol was assayed with current nethod,all three antioxidants inhibittede the peroxi-dation of linoleic acid in direct proportional to to their concention. In conclusion, the the antioxida-tive activity of hydrophorbic antioxildants could be assayed rapidly with current method.
