    Title: 蟾酥複方之抗炎鎮痛及急性肝毒性研究
    Evaluate Anti-innanmzatory, Analgesic effect and Hepatotoxicity on Bofonis herbal Preparation

    Abstract: BUFA1是由蟾酥(Bufonis Venenum)、青黛(Indigo Naturalis)、黃芩(Scutellariae Radix) 和珍珠末(Margarita) 按重量以1:9:9:5的比例所組成的複方。大白鼠在餵食100mg/kg的劑量後,能明顯的減緩Carrageenin及Serotonin所引發的下肢足蹠水腫(Hind-paw edema)。大白鼠在去除腎上腺之後,BUFA1則無抗水腫的作用,顯示BUFA1抗水腫效用與腎上腺有關。另外在餵食相同劑量下,BUFA1能減少小白鼠腹腔注射醋酸(0.1m/10g)後引起的後肢伸張(Writhing Response)次數。然而,對於熱板所引起的疼痛則無止痛作用。在毒性分析實驗中發現,大白鼠連續餵食1週的BUFA1(100mg/kg/day)後,其血中白血球(WBC)和紅血球(RBC)的數目以及血紅素(Hemoglobin)的量並不會受到影響,而且肝功能的指標(SGOT,SGPT及Alkaline Phosphatase)也沒有顯著的變化。除此之外,大白鼠在餵食高劑量(15g/kg)的BUFA1後,在24小時之內並無任何一隻大白鼠死亡的現象(n=15)。綜合以上的結果BUFA1能有效的減緩發炎反應,但是不具明顯的鎮痛作用,而且BUFA1是一安全性高的複方。
    BUFAI is a traditional Chinese medical preparation composed of Bofonis Venenum Indigo Naturalls. Scutellarlae Radix and Margarita in the ratlo of 1:9:9:5 Oral admlnstration of BUFA1 (100 mg/kg)markedly reduced carrageenin and serotonin induced edema in hInd paw of the rat and this anti-inflammatory effect of BUFAI was influencedced by adrenal glands. In the same dosage, BUFAI sighlNcantly attenuated writhing response of the mlce triggered by (i.p.) acetic acid. However, it falled to relieve the pain caused by hot plate. There were no signiflcant changes of blood cell components, including red blood cell, white blood cell and hemoglobin content, as well as liver function tests (SGOT, SGPT and Alkaline phosphatase activity) in the rat after feeding BUFAI (100 mg/kg)fora week. In addition, the rat can tolerate BUFAI as high as 150-fold dosage used for antl-inflammation. In conclusion, BUFAI exhibits an anti-inflammatory effect that is influenced by adrenal glands. BUFAI has no significant analgesic actlon and it appears to be safe.
