Abstract:	本研究的目的在選殖白色來亨雞之端粒反轉錄瓷 (telomerase reverse transcriptase, TERT) 基因並進行 DNA 序列分析，期能發展延長家禽細胞體外培養的技術平台。試驗應用顯微抽取技術，自孵化16 hr的白色來亨雞蛋取得原腸期雞胚並萃取總RNA樣品，經過反轉錄-聚合瓷連鎖反應 (reverse transcription-polymerase chain reaction, RT-PCR) 合成互補 DNA (cDNA)，再設計特異引子進行 PCR以選殖雞的TERT。試驗結果顯示，經過PCR後獲得一長度 4.7 kb的產物，該產物經膠體回收並選殖入TOPO載體進行 T-A cloning，經過 DNA 序列分析與 Basic Local Alignment SearchTool (BLAST) 比對，結果與 NCBI 基因庫中編號 AY505015 序列具有最高之相似性 (99%)，確定為雞的TERT基因。此一 TERT 基因的構築物將可供基因轉殖與表現分析研究之用。
