    Abstract: Fluoroquinolone是屬於quinolone系的抗菌劑,對革蘭陰性菌具廣大之抗菌作用。在眼科的使用上,是用於治療眼睛的膿性發炎症狀;一般所使用的劑型"眼用滴劑"作用期相當短,為了能提供並維持作用部位較長時間的適當藥物濃度,我們已發展出供眼睛投與局部作用之norfloxacin微脂粒藥物處方;本計畫中,我們擬繼續以ofloxacin、enoxacin與ciprofloxacin為模式藥物,將上述藥物包埋於磷脂質小胞中形成微脂粒,隨後,針對脂質組成與其他之添加成分對微脂粒的形態、粒子大小、安定性與包埋率進行研究,同時,感溫親水膠體則另行添加於fluoroquinolone微脂粒眼用溶液中,微脂粒的粒徑將藉助N4庫爾特粒徑分析儀來檢測,微脂粒與眼用製劑的理化性質是以熱分析與組織物性測定儀來分析,安定性的評估則採用加速安定性試驗,微脂粒型Fluoroquinolone之藥物釋出與豬眼睛之經眼角膜藥物輸移則以體外實驗進行研究,而後,微脂粒與眼角膜間之交互作用再以熱分析來探討。上述實驗的結果,將有助於闡明Fluoroquinolone 經角膜傳輸時的微脂粒作用機制。
