    Title: 中草藥對面皰之抑制功效與機轉之探討
    Functional evaluation and mechanism studies of the anti-acne activities of Chinese herbal extracts

    Abstract: 在臨床上面皰治療方式多以使用抗生素或醫學美容方面著手,對身體產生副作用多,易導致細菌的抗藥性以及皮膚的過敏現象。因此本研究中主要以紫草、連翹、仙鶴草、五倍子、蒲公英、川芎、天門冬及訶子八種中草藥,分別探討不同之抗氧化力,以及利用濾紙過散試驗(Paper disc diffusion method)對痤瘡桿菌之抑菌試驗,並將中草藥應用於化粧品配方中評估對預防面皰之生成及抑制作用。
    經由不同抗氧化試驗之評估,在TEAC試驗和pUC119 DNA經‧OH傷害具有保護效果分別以仙鶴草、五倍子及訶子(TEAC之SC50分別為0.022、0.001、0.004mg/ml;DNA保護之SC50分別為19.43、1.76、9.71mg/ml)具有較高的清除能力;在DPPH試驗方面,則以連翹、五倍子及訶子(SC50分別為0.121、0.003、0.012mg/ml)有較高的活性;清除一氧化氮之生成則以紫草、五倍子及訶子(SC50分別為0.075、0.018、0.088mg/ml)之活性較高;pUC119 DNA經ONOO-傷害以紫草具有最強的保護效果,其次則為五倍子及川芎(SC50分別為9.89、19.67mg/ml)。另外在抑菌測試方面,紫草、五倍子及訶子對於抑制痤瘡桿菌生成的能力較強。而在人體試驗之評估,無論對於減少porphyrin之生成,亦或是對於發炎面皰之改善,紫草、連翹、仙鶴草、五倍子、蒲公英及訶子皆能達到明顯改善的作用。因此中草藥對於面皰之活性於體外及體內評估上,呈現出具有開發為化粧品配方之能力。
    Anti-acne treatments are based on the use of antibiotics. However, the increasing antibiotic resistance and the skin irritation have led to the development of alternative therapies such as traditional herbal drugs. In this study, eight kinds of Chinese herbal were employed, they were Lithospermum erythrorhizon SIEB Et ZUCC, Forsythia suspensa (Thunb) Vah.l, Agrimonia pilosa Ledeb, Galla Chinensis, Taraxacum formosanum Kitamura, Ligusticum Chuanxiong, Asparagus cochinchinensis and Terminalia chebula Retz. The results obtained from different anti-oxidative and paper disc diffusion methods were used to analyze the Propionibacterium acnes inhibiting ability, and then apply the Chinese herbal in cosmetic formula to evaluate the prevention and inhibition of cosmetic formulations for the anti-acne.
    By different anti-oxidant activities assay, Agrimonia pilosa Ledeb, Galla Chinensis and Terminalia chebula Retz scavenged ABTS‧+ free radical (SC50=0.022, 0.001, 0.004mg/ml, respectively)and protecting for hydroxyl radical induced pUC119 DNA damage(SC50=19.43, 1.76, 9.71mg/ml, respectively)have highest activity. Forsythia suspensa (Thun) Vah.l, Galla Chinensis and Terminalia chebula Retz scavenged DPPH free radical (SC50=0.121, 0.003, 0.012mg/ml, respectively)have higher activity. Lithospermum erythrorhizon SIEB Et ZUCC, Galla Chinensis and Terminalia chebula Retz scavenged NO production(SC50=0.075, 0.018, 0.088mg/ml, respectively)have better result. Lithospermum erythrorhizon SIEB Et ZUCC show the strongest protection result in protecting pUC119 DNA is damage by peroxynitrite, the second are Galla Chinensis and Ligusticum Chuanxiong.
    In the anti-acne activity test, Lithospermum erythrorhizon SIEB Et ZUCC, Galla Chinensis and Terminalia chebula Retz showed have strong inhibitory effects for Propionibacterium acnes. In vivo, Lithospermum erythrorhizon SIEB Et ZUCC, Forsythia suspensa (Thunb) Vah.l, Agrimonia pilosa Ledeb, Galla Chinensis, Taraxacum formosanum Kitamura and Terminalia chebula Retz were significantly for reducing porphyrin, and also can inhibition acne inflammatory. Therefore, the results from the evaluations of the anti-acne activities in vitro and in vivo for Chinese herbal extracts have shown the potential application in the cosmetic formula in the future.
