    Title: 不同萃取方法對豆類萃取液抗老化能力之影響
    Effect of Different Extraction Methods on Anti-aging Activities of Bean Extracts

    Keywords: 本研究將紅豆(Vigna angularis)、黑豆(Glycine max)、黃豆(Glycine max)、綠豆(Vigna radiata)及花豆(Phaseolus coccineus),分別以冷水浸泡萃取(編號BSE01~05)及超音波振盪萃取(編號BSE06~10),進行對基質金屬蛋白酶(matrix metalloproteinases, MMPs)活性抑制分析及抑菌能力試驗,經分析結果後,發現編號BSE01、BSE05及BSE07,處理3T3細胞培養液後,在5.0~10.0%濃度下反應48小時,對MMP-2及MMP-9抑制活性作用較為顯著;編號BSE01~10對大腸桿菌、綠膿桿菌及金黃色葡萄球菌均無任何的抗菌作用。 為了評估上述3種豆類萃取液(BSE01、BSE05及BSE07)抑制MMPs活性是否有協同作用(synergistic),以等量體積組合為複方萃取液(編號BSE11~13)進行MMPs活性抑制分析,實驗結果顯示,發現編號BSE11(BSE01、BSE05),在5.0~10.0%濃度下,反應24及48小時,對MMP-2及MMP-9都有明顯抑制活性作用。最後將編號BSE11添加調製至基底乳霜中,再進行人體皮膚測試,分析結果顯示,12位受測者使用乳霜4週後,有10位受測者眼周附近皺紋有明顯改善的現象,且12位受測者皆無皮膚刺激過敏反應。 綜合上述實驗結果,複方豆類萃取液可有效降低基質金屬蛋白酶活性,對皮膚具有良好的抗皺能力,能夠開發成為抗老化化粧品原料。

    Abstract: In this study, Vigna angularis, Glycine max, Glycine max, Vigna radiata and Phaseolus coccineus were extracted by water immersion (BSE01~05) and ultrasonic extraction (BSE06~10). Ten extracts obtained from different extraction methods were analyzed by anti-MMP zymography for anti-aging investigation and disc diffusion method for anti-bacterial assay.
    After the zymography analysis, the extracts, including BSE01, BSE05 and BSE07,were treated with 3T3 cell culture exhibited marked inhibitory capacities on MMP-2 and MMP-9 at the concentration of 5.0~10.0% in 48 h. The results from the anti-bacterial assay demonstrated that the extracts of BSE01~10 have no effects on the growth of Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus.
    To evaluate the synergistic activities of the above-mentioned three extracts solutions (BSE01, BSE05 and BSE07), the new three combined extracts (BSE11~13) were prepared by mixing with the equal amount of extracts (BSE01, BSE05 and BSE07) for anti-MMPs zymography. According to the zymography results, the most effective anti-aging extract, BSE11 (the mixture of BSE01 and BSE05) showed the significant suppression effect on MMP-2 and MMP-9 at the concentration of 5.0~10.0% in 24~48 h.
    Finally, the BSE11 extract was added to make the skincare cream for its efficacy assessment on the subjects’ skin. The data from the efficacy testing demonstrated that the BSE11 cream applied to the 12 subjects’ faces for one month displayed obvious improvements on periorbital wrinkles of 10 subjects. Additionally, the BSE11 cream had no irritative effects on the subjects’ faces.
    The above experimental results revealed that the BSE11 extract (the mixture of BSE01 and BSE05) possessed anti-MMP activity and anti-wrinkle ability, and it could be used for the development of anti-aging cosmetic ingredients.
