    Abstract: 在重視生命權的理念下,研發取代動物的方法已成世界趨勢。連細胞培養,因需胚胎牛的血清也遭受批評。但取代不用生命方法一直不能突破。在本研究中我們有新的方法。生物是由化學物質經各種化學鍵結組成的。氫鍵是生命化學鍵結很重要的鍵結,待測物質只要會破壞氫鍵就會破壞結構就具有毒性,對細胞及動物等生命體造成傷害。 因此我們研發生物液膜為合成具有豐富氫鍵的成份。當加入化學物質造成生物液膜氫鍵被破壞時,會由透明變成混濁,檢測其液態核磁共振會改變氫光譜,此化學物就有結構毒。此待測藥物會破壞結構,有非選擇性的毒性,是dirtay骯髒的藥物,加到人體內就會破壞組織有毒性,其在製造使用上就要非常小心。 去年度計畫主要探討本試劑用在化妝品原料及化學物對動物眼睛刺激正確率並與EYTEX試劑比較。結果顯示有近8成以上正確率。 本年度申請之計畫 ,主要為研究本結構毒試劑與動物急毒性LD50數據和細胞毒性IC50數據相關性。 我們要檢測更多的化妝品原料及添加化妝品藥物及各類化學藥品及溶劑,並結合更多的核磁共振圖譜 ,確認本試劑的原理及正確率,以便讓本結構毒性試劑有機會成為國際上各大化妝品廠、藥廠及各實驗室使用。 讓台灣的生技產業能突破應用到國際上
    Under the idea of emphasis on the right of life, replacing animals in the experiment by R & D has become a world trend. Even using embryonic bovine serum in cell culture is criticized. However, the method of not using life in experiment has not made any breakthrough. In this research, we have a new approach. Life is composed of various chemical bonds of chemical substances, and hydrogen bond is a very important one in the chemical bonds of life. Any test substance that is able to destroy hydrogen is able to destroy structure and toxic and may cause damages to cells and animal life. Therefore, we developed a biological liquid membrane that can synthesize rich ingredients of hydrogen bond. When the membrane is destroyed by joining a chemical substance, it will become turbid from transparent. Besides, through the test of its liquid nuclear magnetic resonance (NMR), it will change the hydrogen spectrum, then the chemical substance is a structurally toxic. Such test drug may cause damages to structure and is not selective toxicity and dirty drug that is toxic and able to destroy tissue that we should be very careful in manufacturing and using. Last year’s project aimed to explore the accuracy rates of using reagent in cosmetic raw materials and the chemical substance’s irritancy to the animal eyes and compare it with another reagent EYTEX. The experiment proved that the accuracy rate was near 80%. The project we applied in this year aims to conduct research on structural toxicity reagent and the relationship between the data of animal acute toxicity LD50 and the data of cytotoxicity IC50. We plans to test more cosmetic raw materials, additive cosmetic drugs, various chemical drugs and solvents. In addition, we plan to combine more NMR spectra to confirm the principle and accuracy rates of the reagent. It is hoped that the structural toxicity reagent could be used by well-known international cosmetics factories, pharmaceutical factories and various laboratories that Taiwan’s biotech industries may make a breakthrough and their technology could be used internationally.
