|摘要: ||黃花蜜菜(Wedelia chinensis Merr.)是台灣用來預防及治療肝臟疾患常用民間藥材，具有顯著保護肝臟、腎臟的功效。本研究探討黃花蜜菜(Wedelia chinensis Merr.)酒精萃取粗萃物與其區段萃取物之抗氧化與酪胺酸酶活性抑制效能，並配合高效能液相層析儀鑑定與定量其所含之化學成分，抗氧化實驗結果顯示成分方面，水層萃取物之第II與III區段及EA層萃取物之第IV 與V區段在DPPH自由基清除能力測定、 TEAC 總抗氧化能力、亞鐵螯合能力測定、還原力及微脂粒脂質過氧化抑制能力測定等有較顯著效果；而總多酚含量依序為水層II > EA層V > 水層III > EA層IV，每克萃取物分別相當於508.26 mg、198.19 mg、184.26 mg及161.62 mg之沒食子酸(Gallic acid )；總黃酮含量依序水層II > EA層V > 水層III > EA層IV，每克萃取物分別相當於482.18 mg、359.33 mg、173.47 mg及143.63 mg之芸香素(Rutin )；另外，在酪胺酸酶活性抑制試驗發現其抑制率順序為水層II > EA層V > 水層III>EA層IV在200 μg/mL時抑制率分別為81.7%、88.1%、80.3%及78.8%；HPLC分析亦發現這些區段內含有 Wedelolactone、 Luteolin、 Apigenin、 3,4-Dicaffeoyl-quinic acid及 Quercetin 3-O-β-glucoside等藥理活性成分。綜合所述黃花蜜菜為具有抗氧化與美白潛力之中草藥素材，應可應用於抗老化或美白化妝品。|
Wedelia chinensis, a traditional medicine, has been widely used to protect against the diseases of liver and kidney in Taiwan. We investigated the antioxidant and antityrosinase effects of ethanol extract Wedelia chinensis. The crude ethanol extract was further divided into ethyl acetate (EtOAc) and water fractions by a liquid-liquid partition. In addition, the EtOAc sections and the water sections were further fractionated by silica gel 60 and Diaion HP 20 columns into twelve sections (E1-E5 and W1-W4). Then, we compared the anti-oxidative and anti-melanogensis effects of each section. In the present study, we analyzed the total antioxidant capacity of Wedelia chinensis by the DPPH radical-scavenging, TEAC, reducing power, chelating ferrous ion, and the inhibition of lipid peroxidation. According to the results, the aqueous extracts (W-II&W-III) and EtOAc extracts (EA-IV&EA-V) demonstrated the beat antioxidant activity.The total phenol contents of W-II, EA-V, W-III and EA-IV are 508.26, 198.19, 184.26 and 161.62 mg Gallic acid/g, respectively and The total flavonoid contents of W-II, EA-V, W-III and EA-IV are 482.18, 359.33, 173.47 and 143.63 mg rutin/g, respectively. Moreover, the inhibition activities of tyrosinase on W-II, EA-V, W-III and EA-IV at 200�g/ml are 81.7%, 88.1%, 80.3% and 78.8%, respectively. The high-performance liquid chromatography analysis found that these sections contain wedelolactone, luteolin, apigenin and quercetin 3-O-beta-glucoside and other active ingredients. In conclusion, Wedelia chinensis had excellent effects on antioxidant and antityrosinase activity and maybe applied on antioxidant and antityrosinase products in the future.
