    摘要: 本研究使用離子液體當做液液萃取之萃取劑,應用於快速萃取與濃縮TDI 暴露之大鼠尿液中的芳香胺代謝物:2,4toluene diamine (2,4-TDA) 與2,6-toluene diamine (2,6-TDA),進而使用液相層析質譜儀進行分析。
    同時對於可能影響萃取與濃縮效率的因子進行探討,這些因子包括離子液體的種類與其體積、分散溶液的種類與其體積、樣品溶液的酸鹼值以及樣品鹽類的影響。經過實驗後,最佳的萃取條件是使用經稀釋4 倍的大鼠尿液樣品4 mL , 將樣品pH 值調整為12 後, 加入250 μL 的 1-butyl-3 methylimidazolium hexafluorophosphate([C4mim][PF6])離子液體、
    750 μL甲醇分散溶劑進行萃取;此時溶液形成混濁狀態。然後將樣品放入超音波震盪器中萃取50 分鐘,之後取出萃取後的離子溶液,再添加相同體
    積之含內標準品的甲醇溶液回溶後,注入LC/MS中分析。而使用離子液體萃取TDAs溶液,可將2,4-TDA的萃取效率達到3.5 倍與2,6-TDA的萃取率
    達到3 倍以上。另外,因為萃取過程中只需少量的[C4mim][PF6],因此它也是一種綠色溶劑取代品。
    In this study, the use of ionic liquids as liquid-liquid extraction of extractant, applied to the aromatic amine metabolites (2,4-toluene
    diamine,2,4-TDA and 2,6-toluene diamine, 2,6-TDA) in the rats urine
    for the rapid and enrichment extraction.
    Both factors may affect the efficiency of extraction and
    enrichment discussed, these factors include the type of ionic liquid to its volume, dispersing the impact of the type of the solution, its
    volume, the pH of the sample solution and the
    sample salts. After the experiment, the best extraction conditions with diluted 4 times the rat urine samples of 4 mL, the sample pH was adjusted to 12, adding 250 μL of 1-butyl-3-methylimidazolium hexafluorophosphate ([C4mim][PF6]) ionic liquid and 750 μL methanol as a dispersed solvent in extraction. The sample was then placed in ultrasonic
    vibrator extracted 50 minutes. After xtraction, [C4mim][PF6]
    was removed from solution, and then add the same volume of methanol including internal standard. The mixture was injected into liquid
    chromatography mass spectrometer for analysis. The result showed that the
    extraction efficiencies were improved to 3.0 times higher by use the ionic liquid than organic solvent. The few usage of [C4mim][PF6] is a green solvent
    alternative for extraction.
