    Abstract: 許多環境的物理或化學因子會導致DNA發生變異而致使細胞發生突變,UV-B 是最容易對皮膚造成傷害而使細胞DNA發生突變,也是皮膚癌的重要起因。核?酸切除修補為哺乳類動物細胞移除、修復紫外線及許多常見突變原所造成的DNA損傷之主要修補方式。許多文獻報導中指出非黑色素細胞皮膚癌的起因與核?酸切除修補活性的降低有關。本計畫以蘆薈大黃素(Aloe-emodin)作用於人類皮膚癌細胞株(SCC)和人類皮膚角質化細胞株(HaCaT)後,透過MTS分析、觀察細胞型態之變化測定,來評估經不同濃度的蘆薈大黃素後,SCC和HaCaT不同的影響與差異,並探討蘆薈大黃素增強DNA修補及調節核?酸切除修補的基因表現,進而增強DNA的修補能力相關之機轉,而有預防紫外線引起之皮膚癌作用,並期望能開發有效及專一性的皮膚癌預防及治療上之藥物。
