Effect of Different alcohols on the Formation of Arbutin liposomes

|摘要: ||最近在這一領域的進展，包括乙醇脂質體(Ethosomes)與PG-Liposome的發展。乙醇脂質體通常是含2~5%磷脂質，20~45%乙醇和水，因為乙醇的含量較高而被稱為乙醇脂質體。而PG-Liposome主要由磷脂質、PG和水所組成。本實驗的目的在於結合微脂粒的技術，進行熊果素包覆，將評估添加不同醇類及比例所得微脂粒，製備完成的樣品分別放置在4℃、25℃和45℃環境下，藉由粒徑和粒徑之多分散性指數(P.I.值)的觀察，進行其安定性評估。由實驗結果可得知Ethosomes(20% ethanol、30% ethanol)或PG-Liposomes (10% PG、20% PG) 只要經過擠壓器擠壓的微脂粒，其粒徑、P.I.值都較未擠壓的小，經100 nm濾膜孔徑擠壓後的粒徑最小，經擠壓後其P.I.值皆小於0.21。經100 nm擠壓PG-Liposome(10% PG)其粒徑為131±1；而PG-Liposome(20% PG)粒徑為119±1。Ethosome(20% ethanol)其粒徑為106±1；而Ethosome(30% ethanol)粒徑為71±1，也是此四個配方中粒徑最小的。在所有微脂粒配方中的包埋率皆在79~83 %。經過8小時經皮吸收試驗後，對於添加0.2%熊果素而言，在一般傳統乳液(O/W型乳液)及微脂粒( Ethosome，含30% ethanol )結果顯示，對於熊果素之經皮吸收總量影響不大( P ＞ 0.05 )，不具統計上顯著差異。|
The recent progress in this area, including Ethosomes and PG-Liposome development. Ethosomes is usually 2 to 5% phospholipids, 20 ~ 45% ethanol and water, because higher levels of ethanol, which is known as Ethosomes. The PG-Liposome mainly by phospholipids, PG and water formed. The purpose of this study is the combination of liposome technology to Arbutin coating, will assess the proportion of income to add different alcohols and the liposomes were prepared to complete the samples were placed at 4 ℃、25 ℃ and 45 ℃ environment, diameter particles and the particles diameter by as much as dispersion index (P.I. values) observed that stability into the conduct of its assessment. From the experimental results can be that Ethosomes (20% ethanol、30% ethanol) or PG-Liposomes (10% PG、20% PG) as long as the micro-extrusion through the extrusion device lipid particles, the particles diameter, P.I. values than the non - extrusion a small, membrane pore size by 100 nm extrusion size after the minimum, by extrusion, after the P.I .values were less than 0.21. The 100 nm extrusion PG-Liposome (10% PG) whose particle size was 131 ± 1; while the PG-Liposome (20% PG) particle size was 119 ± 1. Ethosome (20% ethanol) whose particle size was 106 ± 1; while Ethosome (30% ethanol) particle size was 71 ± 1, is the size of the smallest in four formulations. The entrapment efficiency in all liposome formulations varied between 79% and 83%. After eight hours after the percutaneous absorption test, 0.2% for arbutin, the conventional emulsion in the (O / W emulsions) and liposomes(Ethosome, with 30% ethanol) showed a Arbutin percutaneous absorption did not affect the total Large (P> 0.05), not statistically significant.
