    摘要: 雷公藤 (Tripterygium wilfordii Hook F.) 是一種衛矛科的攀緣性藤本植物,可作為中草藥,治療類風濕性關節炎、全身紅斑性狼瘡,並有抗發炎、免疫抑制、抗腫瘤等幼纂A是具有開發潛力之中草藥。因此,我們利用組織培養來誘導雷公藤之癒傷組織,探討癒傷組織在不同溫度、光度、培養基下的影響,希望建立一套雷公藤組織培養的系統,有助於研究及復育雷公藤。本研究以雷公藤的嫩葉和嫩莖作為外植體,培養於 MSB1D0.2 (基礎 MS 培養基添加 1 mg/l BAP 及 0.2 mg/l 2,4-D) 及 MT2B1D0.5 (基礎 MS 培養基添加 1 mg/l BAP 和 0.5 mg/l 2,4-D 和椰子水) 培養基中,誘導癒傷組織之形成,探討不同光度、培養基對誘導癒傷組織之影響,再以癒傷組織作為外植體來探討不同轉速之懸浮培養以及誘導長根之影響。初步結果顯示雷公藤嫩葉和嫩莖的外植體均可誘導癒傷組織,嫩莖與嫩葉於 MT2B1D0.5 培養基誘導癒傷組織較佳,誘導率分別為 81 % 及 79 %;而在黑暗培養下,對嫩莖誘導癒傷組織之誘導率較佳,達 73 %;在懸浮培養中,發現癒傷組織在轉速 100 rpm 培養下較好,重量增加百分比為 85 %;在溫度方面,癒傷組織在 28 ℃ 下生長較佳,重量增加百分比為 81 %;在光度方面,白色癒傷組織在 700 lux 下易變為綠色且重量增加百分比為 92 %;在長根的誘導實驗中,以 1/2 MS 添加 0.1 mg/l NAA 誘導癒傷組織發根率達 60 %,發根數量從 2 至 14 根不等,根長最長為 1.75 公分。另外以帶節的莖段為外植體,經培養後有 93 % 可以長出幼芽,而芽長最長為 1.32 公分;再將芽體移植到長根的培養基中,可以使 87 % 的芽體發根,發根數量從 8 至 15 根不等,根長最長為 2.13 公分;可形成一株小植株。
    Tripterygium wilfordii Hook F. (TWHF) is a celastraceae vine climber, and is a Chinese herb used to treat rheumatoid arthritis and systemic lupus erythematosus, and functions as an anti-inflammatory, immunosuppressor and antitumor. The focus of this study is to establish the technique in tissue culture of TWHF, and attempt to get the undifferentiated calli and induce shoots, roots, and even a whole plant. Stems and leaves of TWHF were cultivated in MSB1D0.2 (MS medium with 1 mg/l BAP and 0.2 mg/l 2,4-D) and MT2B1D0.5 (MS medium with 1 mg/l BAP and 0.5 mg/l 2,4-D and coconut water) media to induce calli. These were then transplanted into suspension culture at different rotation rates and cultivated at different temperatures, light and media conditions. Recent results showed the stem and leaf explants induced calli better at MT2B1D0.5 medium with the ratio of 81 % and 79 % respectively. Calli grew better at 100 rpm, 28 ℃ and dark circumstances with the increased ratio of weight of 85 %, 81 % and 73 % respectively. White calli turned green and grew better at 700 lux with an increased ratio of weight of 92 %. The medium with 0.1 mg/l NAA induced roots with the ratio of 60 %, the root number varied from 2 to 14, and the most length of root was 1.75 cm. 93 % of buds were induced from nodes, the most length of bud was 1.32 cm. 87 % of buds developed a plantlet in 1/2 MS medium with 0.1 mg/l NAA, the root number varied from 8 to 15, and the most length of root was 2.13 cm.
